摘 要:目的 为提高血浆醋氯芬酸的分离效果 ,对高效液相色谱 (HPLC)测定方法进行改进。方法 血浆中醋氯芬酸经盐酸酸化后用乙醚萃取浓缩 ,以Hypersil-C18(4 6mm× 2 5 0mm ,10 μm)为色谱柱 ;以甲醇∶水∶冰醋酸 (70∶30∶0 5 ,V/V为流动相 ,流速为 1 5ml/min ;检测波长为 2 76nm。结果 醋氯芬酸浓度在 0 1~ 10 0 μg/ml范围内线性关系良好 (r=0 996 1) ;最低检出限为 0 1μg/ml;平均回收率为 (10 1 17± 1 17) % ,日内、日间精密度分别为 (6 0 4±2 4 5 ) %和 (5 0 1± 1 39) %。结论 改进后的方法简便、准确可靠、灵敏 ,适用于人血浆醋氯芬酸浓度的测定和药代动力学研究。Objective A modified method based on high performance liquid chromatography was developed for the determination of aceclofenac in human plasma.Methods Aceclofenac was extracted from plasma with diethyl ether.The analytical column was packetd with Hypersil-C 18(4.6mm×250mm,10μ).The mobile phase was methanol-water-glacial acetic acid (73∶30∶0.5,V/V) and the flow rate was 1.5ml/min.The UV detection wavelength was 276nm.Results The calibration curve was liner in the range from 0.1 to 100μ/ml (r=0.9961),with determination limit of 0.1μg/ml and the average recovery of (101.17±1.17)%.The intra-day RSD and inter-day RSD were (6.04±2.45)% and (5.01±1 39)%,respectively.Conclusion The method was simple,rapid, accurate and can be used to determine the aeclofenac concentration in plasma and for pharmacokinetics study.
