保肝类中药复方对肝星形细胞活化及细胞外基质表达的影响

摘　　要：目的　观察保肝类中药复方防治CCl4 所致大鼠肝纤维化模型的疗效 ,并探讨其作用机制。　方法　整体实验 ,采用常规切片、HE、Mallory染色和免疫组织化学ABC法检测Ⅲ型胶原和层黏连蛋白 ;离体实验 ,采用肝星形细胞分离培养 ,检测Ⅲ型胶原和层黏连蛋白 ,并经图像分析系统作定量分析。　结果　保肝类中药复方能减少肝血窦及纤维隔内肌样成纤维细胞的数量 ,抑制纤维隔的纤维组织增生 ,并抑制活化的肝星形细胞分泌Ⅲ型胶原和层黏连蛋白。　结论　保肝类中药复方防治肝纤维化实验疗效明显 ,其抗肝纤维化作用机理与抑制细胞外基质的合成和促进其降解有关。Objective To observe effect of Baoganleifufang on hepatic fibrosis model with CCl-4 and to discuss its mechanism.Methods In vivo, detect collagen Ⅲ and LN by routine section; HE staining; Mallory staining and immunohistochemistry ABC and make quantitive analysis by image analysis system.; in vitro, separate and culture HSC to detect collagenⅢ and LN by routine section and then quantitive analysis is made by image analysis system. Results Baoganleifufang can decrease quantity of muscle-like fibroblast(MFB) in hepatic sinusoid and fiber diazoma and depress the proliferation of fibrous tissue in fiber diazoma. Baoganleifufang can prevent activation of HSC and inhibit it from excreting collagenⅢ and LN.Conclusion Baoganleifufang has significant therapeutics effect on hepatic fibrosis. Its mechanism may be related to depressing synthesis of ECM and accelerating degradation of ECM.
