摘 要:目的 研究羊膜、结膜在光镜、超微结构、免疫组织化学上的异同点。方法 新鲜羊膜、甘油冷冻保存羊膜及结膜 ,分别做HE染色、透射电镜、层粘连蛋白 (LN)及IV型胶原 (CIV)的免疫组织化学染色。新鲜结膜及结膜下组织铺片后 ,分别染色观察杯状细胞及胶原纤维和弹性纤维。结果 电镜下 ,羊膜与结膜基底膜的超微结构相似 ,网状层中胶原纤维的直径相同。LN和CIV在羊膜、结膜的基底层染为连续的颗粒状棕色 ,羊膜的着色明显深于结膜。结论 人羊膜和结膜具有超微结构相似的基底膜层。羊膜与结膜的基底膜具有共同的LN和CIV 。Objective To compare the similarities and differences of structure,ultrastructure and immunoreactivity between amnion(AM) and conjunctiva. Methods Fresh AM,glycerin cryopreserved AM and normal conjunctiva were studied by HE staining under the transmission electron microscope (TEM).Conjunctiva tissue from six eyes was studied by the whole mount.1% alcian blue was used to observe the goblet cells,and Weigert and Van Gieson fluid were used for the staining of collagenous fiber and elastic fiber.Human AM and conjunctiva slices embedded in paraffin were double stained for Collagen IV(C IV) and laminin (LN). Results Under light microscope,AM consisted of three layers including the inner layer with a single layer of columnar cells fixing firmly to an underlying layer of basement membrane,the loose outer layer without vascular and a multilayer conjunctiva epithelium.Lots of microvilli could be seen on the apical surface of AM cells,and at the base,cell pedicels extended into the basement membrane in podocyte fashion.Gap junction and desmosome between adjacent cells,abundant collagen could been seen under the basement membrane under TEM.In whole mount,the numbers of goblet cells in lower-nasal quadrant was the highest among four quadrants.There were abundant collagen and elastic fiber in the Tenon’s fasia,and in the basement membrane of AM and conjunctiva,consecutive granule palm staining was evident.The staining of AM was obviously deeper than that of conjunctiva. Conclusion Human AM and conjunctiva have the same ultrastructure of the basement membrane.The basement membrane of AM and conjunctiva shares the same component of LN and CIV.AM might be useful as a replacement for the conjunctival basement membrane.
