日本血吸虫组织蛋白酶B肽链内切酶基因的获得及结构与功能分析

摘　　要：目的　从日本血吸虫 (Schistosomajaponicum ,Sj)中国大陆株 (chinesemainlandstrain)成虫cDNA文库中获得日本血吸虫新基因 -组织蛋白酶B内切酶基因 (CathepsinBendopeptidase) ,利用生物信息学技术对其结构和功能进行分析和预测。方法　从已构建好的日本血吸虫成虫cDNA文库中挑取重组阳性克隆进行测序 ,对部分序列进行步移法测序获取全长cDNA ,并进行生物信息学分析和登录 ;使用NCBI,SAPS ,TMHMM ,Signal,PsortII,Protscale和Scanprosite等软件对新基因的结构和功能进行分析和预测。结果　对 382个阳性克隆进行测序 ,获得部分新基因 ,其中包括组织蛋白酶B内切酶基因(AY2 2 6 984 )。利用Internet在线分析和相关生物信息学软件对新基因的结构和功能进行了分析和预测。结论　日本血吸虫组织蛋白酶B内切酶基因所编码蛋白为一跨膜蛋白 ,含有半胱胺酸蛋白酶结构域 ,可能对宿主血红蛋白具有降解作用 ,从而在日本血吸虫生长。To obtain genes encoding cathepsin B peptidase from cDNA library of the adult S.japonicum (Chinese strain) and to analyze the structure and function of this organism, clones were selected from the constructed cDNA library and were sequenced. Novel genes were obtained after analysis in GenBank databases from 328 clones, including the cathepsin B endopeptidase gene Cathepsin B endopepyidase of adult S.japonicum was found to be a transmembrane protein with domains of cysteine protease. It suggests that this gene may be related to the function in decomposition of hemoglobulin of host red cells, and play a role in the growth and development of S japonicum.
