摘 要:目的 探讨氨基胍对糖尿病烧伤创面修复的影响及其可能的机制。 方法 SD大鼠随机分为正常对照组、糖尿病对照组和氨基胍干预组。后两组通过腹腔注射链脲佐菌素 (STZ)建立糖尿病模型。STZ注射后 ,干预组予以氨基胍 10 0mg·kg-1·d-1喂养 ,8周后烫伤前为 0时相点 ,三组大鼠给予深Ⅱ度烫伤 ,于伤后 3,7,14 ,2 1d描出创面形状 ,计算创面愈合率 ,观察组织形态学改变 ,并对创面组织糖基化终末产物 (AGEs)含量、糖含量及局部组织释放一氧化氮 (NO)含量进行检测。 结果 与糖尿病对照组比较 ,应用氨基胍后 ,皮肤组织糖含量不降低 ,创面局部炎症反应出现较早 ,坏死组织脱落及上皮匍行提前 ,AGEs含量减少 ,组织释放NO的能力增加 ,创面愈合明显加快。 结论 氨基胍可通过抑制AGEs形成 ,促进NO的合成和释放 ,促进糖尿病烧伤创面愈合。Objective To study the possible effect of aminoguanidine on diabetic burn wound healing. Methods A total of 75 SD rats were randomly divided into non-diabetic control (Group A, n=25), control (Group B) and aminoguanidine application group (Group C). The model was established through injection with streptozotocin (STZ)to diabetes. Group C was feeded with aminoguanidine at 100 mg·kg -1·d -1. Every experimental time point represents five rats from three groups. Eight weeks later, 15 rats were killed for the first experimental time point, and the others inflicted with 20% TBSA of deep partial thickness scalding on the back. The samples from wound sites were harvested at post-burn days (PBD) 3, 7, 14, and 21 to histologically observe the healing process and detect the levels of NO released from local tissues culture medium, the healing rate, the content of advanced glycation end products (AGEs) and the glucose in local tissues. Results In the Group C, the inflammation emerged earlier, necrotic tissues shade in advance, with unchanged glucose of skin tissues but with increased healing rate and concentration of NO and decreased content of AGEs. But the content of glucose had no changes Conclusion Aminoguanidine can improve diabetic burn wound healing through promoting synthesis and release of NO by inhibiting formation of AGEs.
