摘 要:目的:建立用高效液相色谱法测定复合氨基酸胶囊中氨基酸的含量。方法:采用6-氨基喹啉-N-羟基琥珀酰亚胺基氨基甲酸酯(AQC)为衍生剂,与各种氨基酸柱前衍生后,用Waters 2690高效液相色谱仪,AccQ.Tag C18柱(150 mm×3.9 mm,4 μm),以水为溶剂A,Eluent(醋酸盐-磷酸盐缓冲液)稀释液为溶剂B,乙腈为溶剂C进行梯度洗脱,检测波长为248 nm,柱温37℃,进样量10μL。结果:所有氨基酸在37 min内测定完毕,回收率为101.4%-110.2%,RSD 1.1%-4.5%。结论:本法分离度好,快速,简便。可作为产品的质量控制方法。Objective: To establish a method for determination of amino acids in compound amino acid capsules. Method:The amino acids reacted with 6 - aminoquinoly - N - hydroxysuccinimdyl carbamate ( AQC ) respectively and quantitatively and produced corresponding derivatives which were analized by Waters 2690 HPLC. Chromato graphic conditions were AccQ - Tag C18 for amino acids analysis( 150 mm ×3. 9 mm,4μm) ,mobile phase of program elution (A ) water, ( B ) Eluent buffers, (C ) acetonitrile. Detected at 248 nm by UV - detector , column temperature was at 37 o@ ,the injection volume was 10 μL. Results: All kinds of amino acids were completely determined in 37 minutes,and the recoveries were 101. 4% - 110. 2% ,RSD of all amino acids were 1. 1% -4. 5%. Conclusion: The method is simple, accurate and suitable for the determination of amino acids in compound amino acid capsules.
