摘 要:目的 观察大鼠在急性低压缺氧环境暴露后脑皮质和海马细胞凋亡的情况。 方法 采用Wistar大鼠 30只 ,在低压舱内模拟暴露于 75 0 0m高度停留 2 0min ,建立急性缺氧模型。分别在缺氧前及低压缺氧环境暴露后 1d、2d、3d、4d和 5d ,取大鼠脑颞叶皮质和海马组织用流式细胞仪分析脑细胞周期和细胞凋亡率的变化。 结果 大鼠低压缺氧后脑皮质和海马细胞凋亡率明显升高 ,在缺氧后 4d和 3d脑皮质和海马凋亡细胞数分别达到高峰 ,急性缺氧后 3d和 4d脑皮质和海马凋亡细胞数显著高于 1d (P <0 0 1 )。而急性缺氧后脑皮质和海马细胞G1 期细胞呈逐渐减少趋势 ,S期细胞逐渐增加。 结论 75 0 0m模拟高空低压缺氧可导致不同程度的脑皮质和海马细胞凋亡 ,缺血缺氧后的 3d和 4d凋亡细胞率最高 。Objective To observe the dynamic change of apoptotic rate of cells from brain cortex and hippocampus tissue after acute hypobaric hypoxia in rats Methods Thirty Wistar rats were divided into control group and 5 experimental groups The experimentals were exposed to 7500 m altitude in animal hypobaric chamber for 20 min, and brain tissues were taken 1 d, 2 d, 3 d, 4 d and 5 d after exposure for detection of percentage of cells of different stages and rate of apoptosis with flow cytometer Results The rate of apoptotic cell(BAD) from brain tissues of rats increased day by day after hypoxia exposure and the peak value was 3 d after exposure (hippocampus) and 4 d after exposure (cortex) BAD of brain tissue lowered 5 d after exposure but still higher, although non significantly than that of control group The changes of different stage of cells revealed the similar course Conclusions The apoptosis of brain peaked 3 or 4 day after acute hypoxic exposure According to the apoptotic rate, cortex is more sensitive to acute hypoxia than hippocampus
