摘 要:背景:荞麦种子提取物具有降糖、降脂、抗氧化等活性,荞麦花提取的有效成分是否也具有降糖、降脂、抗氧化等作用,尚需进一步的研究。目的:研究荞麦花总黄酮(totalflavonesofbuchwheatflower,TFBF)对实验性2型糖尿病高脂血症大鼠降血糖、血脂等作用,并探讨其降血糖和调节血脂的作用机制。设计:完全随机对照的实验研究。地点、材料和干预:本实验在华北煤炭医学院药理学教研室和华北煤炭医学院附属医院同位素科完成。实验选用SD大鼠60只,清洁级Ⅱ级,体质量(220±20)g,雌雄各半。大鼠被随机分为正常对照组、实验对照组、TFBF0.15,0.30,0.60g/kg剂量治疗组和阳性对照药HB-419组,每组10只。用链脲佐菌素和高脂饲料诱发大鼠2型糖尿病和高脂血症,以TFBF口服治疗12周,剂量分别为0.15,0.30,0.60g/kg。主要观察指标:TFBF对实验性2型糖尿病高脂血症大鼠血中空腹血糖、糖耐量、血清总胆固醇、三酰甘油、低密度脂蛋白胆固醇(lowdensitylipoproteincholesterol,LDL-C)、高密度脂蛋白胆固醇(highdensitylipoproteincholesterol,HDL-C)、动脉粥样硬化指数(atherosclerosisindex,AI)、胰岛素及肝细胞膜胰岛素受体(Insulinreceptors,INS-R)结合力的影响。结果:TFBF治疗组大鼠血中空腹血糖,三酰甘油,LDL-C犤TFBF0.15,0.30,0.BACKGROUND:It has been studied that extract of buckwheat seed has activities of lowering serum glucose,lipids,anti oxidation and so on.But whether the extract of buckwheat flower has the same effects requires further study. OBJECTIVE:To study the effects of total flavones of buckwheat flower(TFBF) on type 2 diabetic and hyperlipidemia in rats on lowering serum glucose,lipids,etc.and to discuss the mechanism of lowering serum glucose and regulating serum lipids. DESIGN:A completely randomized controlled trial. SETTING,PARTICIPANTS and INTERVENTIONS:This study was helped by Department of pharmacology,North China Coal Medical College and Department of isotope,the Affiliated Hospital of North China Coal Medical College.A total of 60 female and male(half half) Sprague Dawley(SD) rats were provided,cleaning gradeⅡ,mass(220±20) g.They were divided into normal control group,experimental control group,TFBF groups of 0.15,0.30,0.60 g/kg and HB 419 group at random and ten in each group.The diabetic and hyperlipidemia models of rats were induced by streptozotocin and high lipid feed,at the same time,TFBF was administered orally at the dosage of 0.15,0.30,0.60 g/kg for 12 weeks. MAIN OUTCOME MEASURES:The combinative effect of TFBF on fasting blood glucose(FBG),glucose tolerance,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL C),high density lipoprotein cholesterol(HDL C),atherosclerosis index(AI),insulin(INS)and insulin receptors of liver cell membranes in type 2 diabetic and hyperlipidemia SD rats' blood was observed. RESULTS:FBG,TG and LDL C in TFBF groups of 0.15,0.30,0.60 g/kg were(8.22±3.04),(7.79±2.68) and(7.04±2.57) mmol/L,respectively;TG was(0.54±0.22),(0.48±0.12) and(0.46±0.20) mmol/L,respectively;LDL C was(1.93±0.26),(1.79±0.22)and(1.72±0.31) mmol/L,respectively,which were lower than that in experimental control group[(11.15±2.80),(0.73±0.15) and(2.47±0.23) mmol/L](t=2.242-6.756, P< 0.05-0.01)]
