摘  要:使用干细胞生长培养基 (SCGM )和RPMI16 4 0培养基 ,设计不同浓度的抗CD3单抗、IL 2和PHA的培养条件 ,从健康成人外周血单个核细胞 (PBMC )中高效扩增细胞因子诱导的自然杀伤细胞 (CINK ) ,并用MTT法检测其细胞毒活性 ,优化CINK细胞的体外扩增技术。用优化的方法扩增 12例不同类型恶性肿瘤患者CINK细胞。结果显示 ,在以SCGM为基础培养基时 ,PBMC经抗CD3单抗、IL 2作用后获得大量增殖 ,在PHA存在时获得最大增殖 (P <0 0 5 ) ,扩增的CINK细胞以CD3 CD5 6 + NK细胞为主 ,扩增细胞的细胞毒活性显著高于相同来源的CIK细胞和CD3AK细胞 。The cytokine-induced natural killer(NK) cells(CINK) from peripheral blood of healthy adults were expanded in stem cell growth medium(SCGM) or RPMI 1640 medium supplemented with anti-CD3 monoclonal antibody, IL-2 and PHA at varying concentrations, and the cytotoxic activity of CINK was determined by MTT assay. By using the optimizing techniques of in vitro expansion, the CINK from 12 cases with various kinds of malignant tumors were expanded. The results showed that CINK expanded with high efficiency cultivated in SCGM suplemented with anti-CD3 monoclonal antibody and IL-2 and the highest degree of proliferation was observed in the presence of PHA. The expanded cells of CINK were mainly the CD3+ CD56+ NK cells and their cytotoxic activities were markedly higher than those of CIK or CD3AK cells against tumor targets(K562 and Raji cell lines). This works would provide an experimental basis for the adoptive immunotherapy to tumors with auto-CINK cells.
