摘 要:采用分离酶(dispase)冷消化法探寻纯净口腔粘膜基底层上皮细胞的原代培养方法,以用于口腔粘膜上皮细胞与钛结合机理及其改善因素的研究。结果表明:dispase的作用部位位于口腔粘膜的基底膜和基底细胞层之间,dispase冷消化法原代培养成功的细胞来自口胶粘膜的基底层,无成纤维细胞成分。结论:用dispase冷消化法可以建立符合临床实际的研究口腔粘膜上皮细胞与钛结合机理及其改善因素的上皮细胞体外培养模型。It is necessary to establish a model of culturing nonnal human ond mucosal hasal cells which ndndced thecharaterishcs in vivo structually and functionally to investigate the mechanism and the imPmeing factors ofthe attahTnent bebeen titanium and epithelium. In this researeh, the enzyme dispase was used to free themucosal epidends from the underlying strom.This yields an epidermal sheet in which no fibroblast cell wasexist.After tmpsinisation with 0.125% topsin for 5 ndnutes, epithelial cells were isolated and were cultured. Histosection exndnation stained with Gomori's and H. E. showed that dispase acts at the dermal - epidermal junchonand the mucosal basal cells is the maority cell type in the cultured cells. The cells grew as amonolayer and could sutwved for at least 2 weeks. It suggests that the culture of human oral mucosal basal cellsmeet with success .
