摘  要:目的 研究中研II号治疗猴艾滋病的作用机理。方法 采用流式细胞仪检测法分析细胞周期 ;WesternBlot检测丝裂原活化蛋白激酶 (ERK2 )活性及电泳迁移率改变检测法 (EMSA)检测转录因子NF κB和AP 1活性。结果 对SIV感染猴模型 ,中研Ⅱ号可阻断HI SIV及HIVgp12 0介导的淋巴细胞功能 ,能抑制ERK2的活性 ,并降低转录因子NF κB和AP 1活性 ,调控HI SIV诱导的CD3/CD4 刺激信号转导通路。结论 中研Ⅱ号调节猴艾滋病T细胞功能可能是通过CD4 共刺激T细胞受体 (TCR)Objective To investigate the mechanism of Zhongyan No.Ⅱ for the treatment of AIDS in the monkeys. Methods Flow cytometry was used to detect and analyze the cell cycle, Western Blot analysis was used to detect the activity of mitogen activated protein kinase (ERK2), and electrophoresis mobility shift assay was used to detect the activities of nuclear transcription factor-kB (NF-kB) and activator protien-1 (AP-1). Results Zhongyan No.Ⅱ could block the lymphocytic function mediated by HI-SIV and HIVgp120, inhibit the activity of ERK2, decrease the activities of NF-kB and AP-1, and regulate and control the CD 3/CD 4 stimulation signal transduction pathway induced by HI-SIV. Conclusion The exertion of the effect of Zhongyan No.Ⅱ for regulating the function of T lymphocytes in the monkeys with AIDS may be through a signal cascade involving CD 4 co-stimulated T cell receptor signal pathway.
