摘  要:目的 研究制作实验性自身免疫性肌炎动物模型的方法。方法 利用豚鼠骨骼肌肌浆球蛋白 (蛋白浓度分别为 6g/L和 3g/L)加完全弗氏佐剂 (含卡介苗 )多次免疫大鼠 ,或联合腹腔注射百日咳杆菌原液 (细菌数为 4 0× 10 10 个 /只 ) ,制成实验性肌炎模型 ,观察其在血清肌酶、血清抗肌球蛋白抗体、肌电图、病理的改变。结果 发现其与人类多发性肌炎 (PM)在临床症状、肌酶、肌电图、病理上的改变有相似之处。结论 可作为研究人类多发性肌炎 (PM )的一个重要手段 。Objective To find out a method for establishing the animal model of experimental autoimmune myositis. Methods The animal model of experimental autoimmune myositis was induced by repeatedly immunizing the rats with guinea-pig skeletal muscle myosin (the concentration was 6 g/L and 3 g/ L, respectively) plus Freund's complete adjuvant (containing BCG vaccine), or giving a celiac injection of the stock solution of Bordetella pertussis (at a dose of 4.0×1010 bacteria of Bordetella pertussis each rat) in addition. The changes in serum muscle enzymes, serum myosin antibodies, electromyography parameters and other pathological respects were observed. Results It was found that the animal model of experimental autoimmune myositis induced by the above-mentioned procedure was similar to human polymyositis concerning the clinical symptoms, serum muscle enzymes, electromyography parameters and other pathological respects. Conclusion The animal model of experimental autoimmune myositis established in this experiment offers an important approach to the study of human polymyositis and is also a good animal model, which can be used to investigate the pathogenesis and treatment of human myositis.
