摘 要:目的观察神经前体细胞植入新生鼠局灶性缺血区内的存活和融合情况。方法应用含表皮生长因子+碱性成纤维生长因子诱导因子的无血清D/F12(1:1)培养获得新生鼠神经前体细胞球并在培养中掺人5-溴-2'-脱氧尿嘧啶标记,免疫酶技术检测nestin标志和分化诱导实验证明所获得的细胞球是神经前体细胞,去脑膜结扎小动脉手术制作新生鼠脑皮层局灶性缺血动物模型,将标记好的神经前体细胞直接注射到缺血损伤灶的边缘区,定期免疫组化染色观察植入细胞的分布、存活和与宿主脑组织的融合情况。结果缺血损伤侧的脑皮层内带标记的细胞量明显要多于实验对照的对侧,植入的神经前体细胞主要向损伤灶的颗粒层聚集,未见远部位迁移,与宿主脑皮层组织有很好的融合。结论大脑皮层缺血性损伤可刺激神经前体细胞发育,提示新生动物缺血性脑损伤疾病有希望通过神经前体细胞移植获得治疗。Objective To investigate the fate of the neural precursor cells transplanted into focal cerebral cortex ischemic boundary zone of newborn rats. Methods A free-serum medium D/F12 (1:1) containing basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) was used to obtain the neural precursor cell spheres and BrdU was incorporated into the neural precursor cells to mark them during the period of culturing in vitro. The neural precursor cells were identified by detecting the neural precursor cell mark nestin with immuno-enzyme techniques and inducing differentiation tests. The neural precursor cells marked with BrdU were transplanted into the cerebral ischemic boundary zones of the newborn rat models which were made by removing the cerebral menings to stop the blood supply for the neop-allium. Living and incorporating of the grafts in the host cerebral cortex were detected at defined time points with the immuno-histochemical technique. Results The grafts marked with BrdU living in the in-juried cerebral cortex are significantly more than that in the opposite intact cerebral cortex as the experiment controls. The grafs mainly focused on the granular layer of the injuried cerebral cortex and not found in remote sites from the lesion, It was watched that the cells marked with BrdU well integrated into the host cerbral cortex tissues. Conclusion The ischemic lesion of cerebral neocontex of rats can activate the neural precursor cells to develop, suggesting that transplantation of neural precursor cells can be used to therapy the cerebral cortex ischemic lesion of newborns.
