摘 要:目的探讨干扰素诱导蛋白10(interferon-inducibleprotein10,IP-10)是否参与干燥综合征唇腺的炎性反应。方法对49例干燥综合征患者行唇腺活检,所得标本进行HE染色,检测并计数4mm2组织中淋巴细胞浸润灶。对其中39例(原发性干燥综合征21例,继发性干燥综合征5例,其他疾病13例)行RT-PCR检测。提取总RNA并反转录成cDNA。根据IP-10cDNA序列设计特异引物。以唇腺标本cDNA为模板,同时加入IP-10和β-actin的特异引物行PCR共扩增。扩增产物电泳后测量IP-10和β-actin相应条带的光密度,计算两者光密度的比值,对标本中IP-10mRNA的表达水平进行半定量分析。对21例唇腺标本(原发性干燥综合征10例,继发性干燥综合征3例,其他疾病8例)行免疫组织化学染色。对11例标本同时完成RT-PCR和免疫组织化学检测。结果干燥综合征患者唇腺标本IP-10与β-actinRT-PCR共扩增后光密度比值为0.329±0.157,而非干燥综合征患者为0.099±0.059,两者相比差异有显著性(P<0.01)。继发性与原发性干燥综合征患者唇腺标本中IP-10的表达差异无显著性。IP-10与β-actin光密度比值与干燥综合征患者唇腺标本中淋巴细胞浸润灶计数呈正相关(r=0.657,P<0.05)。免疫组织化学结果显示,10例原发性干燥综合征唇腺中有8例为阳性(8/10),而继发性干燥综合征3例均?To investigate whether interferon-inducible protein10(IP-10)is involved in the inflammatory process of the labial gland of patients with Sjogren's Syndrome (SS).Methods Forty-nine patients performed labial gland biopsy,the number of lymphocytes in the biopsy tissues was calculated and the IP-10was detected by the methods as following:39biopsied labial tissues were examined by RT-PCR,among them,21were from primary SS,5from secondary SS and13from other diseases.With RT-PCR,the IP-10andβ-actin were co-amplified with specific primers.The gel-fractioned and ethidium bromide amplification products were then analyzed by densitometry.The expression of IP-10was semi -quantificated by IP-10/β-actin ratio.Twenty-one samples were examined by immunohistochemistry with specific goat anti-IP-10antibody,10of them from primary SS,3from secondary SS,8from other diseases.11out of21samples were examined by both RT-PCR and immunohistochemistry.Results The expression of IP-10mRNA was significantly up-regulated in labial glands of patients with SS compared with other diseases(IP-10/β-actin ratio was0.329±0.157vs0.099±0.059,P<0.01).The number of lymphocyte infiltration foci in labial glands of patients with SS correlated to the IP-10/β-actin ratio(r=0.657,P<0.05).Ductal epithelial cells and some of the infiltrating lymphocytes were stained by anti-IP-10antibody by immunohistochemistry in8of the primary SS(8/10),all of the secondary SS(3/3)and one with primary biliary sclerosis(1/8).The expression of IP-10protein detected by immunohistochemistry was consistant with that of mRNA detected by RT-PCR.Conclusions IP-10is abnormally highly expressed in the labial glands of patients with SS and positively relates to the lymphocyte infiltration.It thus suggests chemokine IP-10may be one of the important molecules attracting the lymphocytes to the minor salivary glands to form the lymphocytic foci of Sjogren' s Syndrome.
