摘  要:目的 观察高糖对体外培养的大鼠肾小球系膜细胞(GMC)表面β1整合素表达和细胞外基质(ECM)分泌的影响。方法 应用不同浓度的葡萄糖刺激培养GMC,采用MTT法检测细胞增殖,RT-PCR法检测GMC表面β1整合素的表达,ELISA法检测纤维连接蛋白(FN)、层粘蛋白(LM)的分泌情况。结果 高糖抑制GMC的增殖,促进β1整合素的表达,且与FN和LM的分泌呈显著正相关,并具有浓度依赖性。结论 高血糖作为糖尿病肾病的启动因素可能是通过上调β1整合素mRNA的表达,从而促进ECM成分FN和LM的合成分泌。Objective To observe the effects of high glucose on β1 integrin mRNA expression in cultured glomerular mesangial cells ( GMC). Methods Cultured glomerular mesangial cells were treated with glucose in different concentrations. The proliferation of GMC was measured by MTT, the expression of β1 integrin was estimated by RT-PCR, and the secretion of fibronectin and laminin were measured by ELISA, respectively. Results High glucose inhibited the proliferation of GMC and promoted the expression of β1 integrin, which had a significant correlation with the secretion of fibronectin and laminin. The effect was dose-dependent. Conclusion Hyperglycemia, as an initial factor of diabetic nephropathy, may increase the synthesis and secretion of fibronectin and laminin of the extracellular matrix (ECM) through the up-regulation of β1 integrin mRNA expression.
