摘  要:目的 研究小鼠胚胎心房肌和心室肌细胞自律性水平随胚胎发育不同时期而改变的发育依赖性特点。方法采用全细胞膜片钳技术 ,在电流钳条件下 ,记录了不同胚胎发育阶段小鼠单个心室肌细胞和心房肌细胞的跨膜动作电位。结果 心室肌细胞早期 (9 5~ 10 5d ,earlydevelopmentstage,EDS)、中期 (11~ 14d ,intermediatedevelopmentstage ,IDS)、晚期 (14~ 18 5d ,latedevelopmentstage,LDS)动作电位周期分别为 (172 86± 2 6 15 )ms、(2 4 7 73±15 84 )ms、 (32 6 6 7± 30 10 )ms (P <0 0 5 ) ;中、晚期心房肌细胞动作电位周期分别为 (2 0 4 15± 2 4 14 )ms,(2 2 6 5 3± 2 0 2 8)ms。结论 随着胚胎发育成熟 ,小鼠胚胎心房肌和心室肌细胞的自律性活动逐渐降低。Objective To investigate the characteristics of development-dependent change in autorhythmic activities of single atrial and ventricular myocyte of mouse embryo as the embryo develops. Methods Whole cell patch clamp technique with current being clamped was used to record the action potential of single atrial and ventricular myocyte of mouse at different developmental stages of embryo. Results The duration of active potential of mouse ventricular myocyte at early developmental stage (EDS), intermediate developmental stage (IDS) and late developmental stage (LDS) was (172.86±26.15) ms, (247.73±15.84) ms, (326.67±30.10) ms respectively (P<0.05), and the duration of active potential of mouse atrial myocyte at IDS and LDS was (204.15±24.14) ms and (226.53±20.28) ms respectively. Conclusion The autorhythmic activities of both atrial and ventricular myocytes of mouse embryo are decreased gradually as the embryo develops.
