摘  要:目的 :通过探讨不同给苯剂量、苯作用时间的小鼠反应 ,筛选建立苯诱发小鼠再生障碍性贫血模型的方法。方法 :予雄性CD1小鼠每周三次皮下注射“苯 +玉米油”混合液 ,按给苯剂量不同分为四组 :B1组(0 .5ml/kg)、B2组 (1.0ml/kg)、B3组 (1.5ml/kg)、B4组 (2 .0ml/kg) ,以单纯注射玉米油组 (A组 )为对照 ,观察小鼠一般状况、血象、骨髓象、骨髓有核细胞计数 ,并进行肝、脾和胫骨的组织病理学观察。结果 :与对照组相比 ,B4组在给苯 2 5次后体重和脾脏指数分别下降 18.5 1%和 6 3.86 % (P <0 .0 1) ,白细胞、红细胞、血红蛋白、血小板、网织红细胞绝对值和骨髓有核细胞计数分别下降 74 .97% (P <0 .0 1)、4 1.6 6 % (P <0 .0 1)、30 .96 % (P <0 .0 1)、2 4 .77% (P <0 .0 5 )、4 1.4 7% (P <0 .0 5 )和 6 1.6 1% (P <0 .0 1) ,骨髓涂片检查和病理形态学观察均显示造血细胞减少、脂肪细胞和淋巴细胞等非造血细胞增加 ;综合以上结果提示本实验中苯诱发再障模型建立成功。结论 :予CD1小鼠每周三次皮下注射 2ml/kg体重的苯 ,累计给苯 2 5次 ,共 5 0ml/kg ,初步建立了苯诱发小鼠再生障碍性贫血模型的实验方案。Objective: To establish a model of aplastic anemia induced by benzene in mice. Methods:CD1 male mice were injected subcutaneously with benzene +corn oil three times a week, and randomly divided into four groups according to the dose of benzene administrated: B1 group(0.5 ml/kg),B2 group(1.0 ml/kg ) ,B3 group(1.5 ml/kg) and B4 group(2.0 ml/kg), while A group treated with on ly corn oil as control.The parameters tested were as follows: general conditio n, peripheral blood smear, bone marrow smear,bone marrow nucleated cell count, and histopathology of liver,spleen and tibia. Results:After exposure to 25 benzene doses, compared with A gro up, B4 group mice showed 18.51% body and 63.86% spleen/body weight ratio declin ed separately( P<0.01), and leukocyte(74.97%,P<0.01 ),erythrocyte(41.66%,P<0.01),hemoglobin(30.96%,P <0.01),platelet(24.77%,P<0.05),reticulocyte (41.47%, P <0.05) and bone marrow nucleated cell count (61.61%,P<0.01) we re decreased,the bone marrow smears and the patho-morphological examinations showed that hemato poietic cells were decreased with the increase of the non-hematopoietic cells,s uch as lymphocytes and fat cells; these parameters indicated that the model was set up successfully. Conclusion:An experime ntal aplastic anemia model in mice is established preliminarily by subcutaneous injection of 2 ml/kg benzene. three times a week, summing up to 25 times and 50 ml/kg.
