摘 要:甘薯叶片培养在附加NAA0.01-1.0/ZT0.01-10mg/L的MS培养基上可以100%地诱导产生愈伤组织,NAA1.0/ZT0-0.1mg/L可诱导叶片产生根。经HPLC测定,甘薯叶片内源生长素含量为480ng/g·FW,可能与甘薯叶片易诱导生根有关。在MS培养基上诱导产生的甘薯愈伤组织质地致密、生长缓慢,当继代于高NO_3^-低NH_4^+含量的改良MS培养基上就可转变为疏松、生长迅速的愈伤组织。用疏松愈伤组织酶解出了大量原生质体,并培养再生成了愈伤组织块。This paper reported the callus formation and endogenous phytohormone IAA content of the leaf explants of sweet potato , the growth regulation of callus and protoplast regeneration. The main results were as follow: The frequency of callus formation of leaf explants was 100% on MS medium (NAA 0.01-1.0/ ZT 0.01-10mg/ L) .At the same time, roots could produce on medium (NAA1.0 / ZT 0-0.1mg / L) . The IAA content of the leaf from sweet potato was 480ng / g.F W measured by the HPLC. The texture of callus was fine and close , growthed slowly on MS medium . But the texture of cullus was loose and growthed quickly on improve MS medium ( high NO3-, low NH4+) . Using loose callus, the protoplasts were obtained by enzymatic digestion and regeneration into callus lines .
