摘 要: 研究了以海藻酸钠/壳聚糖/海藻酸钠为囊壁材料的胶囊固定化辣根过氧化物酶的制备及其影响因素,并将胶囊固定化辣根过氧化物酶/乙酰丙酮/H2O2酶促体系用于引发丙稀酰胺的聚合。结果表明,作为囊壁材料之一的壳聚糖的平均分子量以1万左右为宜,以柠檬三酸钠溶液为溶芯剂的溶芯时间控制在2~4min时,固定化率可达60%,相对于游离酶,其相对活力约为60%。胶囊固定化辣根过氧化物酶/乙酰丙酮/H2O2酶促体系可引发丙稀酰胺的聚合。The preparation of the capsule-immobilized horseradish peroxidase(HRP) with alginate-chitosan-alginate as the materials of capsule shell was studied. The polyacrylamide was synthesized by the initiation of the enzyme catalysis system composed of the capsule-immobilized HRP/acetylacetone/hydrogen peroxide. The results show that the alginate with average molecular weight of 1.0×10~4 is suitable for using as the shell material for capsule, and the core-denude time with the sodium tri-citrate as the core- denuding agent should be limited to 2-4 min. The immobilized efficiency of enzyme is about 60% and the relative specific activity of capsule-immobilized enzyme are about 60% respectively. The experimental results of polymerization show that the polymerization of acrylamide could be initiated by the catalysis system of capsule-immobilized HRP. According to the analysis of polymerization mechanism, it is known that the polymerization is initiated by the radicals formed inside of the capsule , which are diffused across the shell of the capsule to the zone of reaction.
