两个地理株青海血蜱基因组DNA差异研究

摘　　要：目的　研究青海血蜱是否有种型差异。方法　分别提取青海血蜱两个地理株的DNA ,选取 8条不同的随机排列碱基顺序的多聚核苷酸单链为引物 ,进行RAPD -PCR扩增反应。将扩增产物制备成DNA图谱 ,比较青海血蜱两个地理株基因组DNA多态性。从DNA水平对青海血蜱两个地理株进行差异分析。结果　青海血蜱两个地理株大部分DNA条带是相同的 ,少数DNA条带不同。结论　两个地理株青海血蜱DNA存在着差异。Objective To study whether two strains of Haemaphysalis qinghaiensis had genomic DNA differences. Methods The genomic DNA was extracted respectively from two strains of Haemaphysalis qinghaiensis,8 primers with different arbitrary single chain polynucleotide sequences were selected,the genomic DNA was amplified by RAPD-PCR.DNA fingerprint maps were made with the amplified products.The polymorphism DNA of the two strains of Haemaphysalis qinghaiensis was compared,and their diversities were analyzed on DNA level. Results Major DNA fragments between two strains were the same,minor DNA fragments between them were different. Conclusion The diversities of genomic DNA between two strains of Haemaphysalis qinghaiensis existed.
