复方盐酸二甲双胍片中盐酸二甲双胍和格列本脲的HPLC测定

摘　　要：建立了HPLC法测定复方盐酸二甲双胍片中盐酸二甲双胍和格列本脲的含量。以双氯芬酸钠为内标,采用HypersilBDS C18柱,流动相为甲醇-3mmol/L十二烷基硫酸钠溶液(含0.1%三乙胺,pH 3.5)(67∶33),流速 1ml/min,检测波长分别为232和300nm。盐酸二甲双胍和格列本脲分别在0.03～0.3mg(r=0.9998)和0.5～4mg(r =0.9999)范围内线性关系良好。方法平均回收率分别为100.1%(RSD 0.22%)和99.9%(RSD 0.36%)。A HPLC method for determination of metformin hydrochloride and glibenclamide in compound metforminhydrochloride tablets was established. Diclofenac sodium was used as the internal standard. A Hypersil BDS C18 column wasused with the mobile phase of methanol-3mmol/L lauryl sodium sulfate solution(containing 0.1% triethylamine, pH 3.5)(67∶33). The flow rate was 1ml/min. The detective wavelengths were 232nm and 300nm, respectively. The calibrationcurves of metformin hydrochloride and glibenclamide were linear in the range of 0.03～0.3mg(r=0.9998) and 0.5～4mg(r =0.9999), respectively. The average recoveries were 100.1%(RSD 0.22%)and 99.9%(RSD 0.36%), respectively.
