益肾活血中药肾康灵干预阿霉素肾病大鼠的药理研究

摘　　要：目的 :通过观察益肾活血中药肾康灵对阿霉素肾病 (AN)大鼠的干预作用及与核因子 (NF -κB)等的关系 ,阐明NS的发病机制及益肾活血中药干预治疗的可能机制。方法 :用阿霉素 (ADR) 5 .5mg/kg诱导大鼠类似于人类微小病变 (MCD)型肾病模型 ,用泼尼松 (C组 )、泼尼松 +肾康灵 (D组 )治疗 ,并设对照组。检测各组治疗前后NF -κΒ及肾脏病理形态学等指标的改变。结果 :AN大鼠在治疗前尿蛋白显著增多 ,Alb显著下降 ,Ch和TG、NF -κB活性显著增强 ,肾脏病理损害明显加重 ;在治疗后 ,治疗组AN大鼠上述指标都明显改善 (P <0 .0 5或P <0 .0 1) ,肾脏病理损害明显减轻。结论 :尾静脉注射ADR造成的人类肾病病理模型 ,其临床表现和实验指标改变类似于中医的“肾虚血瘀”病变 ;益肾活血中药肾康灵能显著改善AN鼠的肾虚血瘀证表现 ;NFObjective:To observe the effects of Shenkangling (a TCM decoction with the effect of benefiting the kidney and promoting the blood circulation) on the Adriamycin-induced nephropathic rats (AN rats),and its relations with the nuclear factors such as Kappa B(NF-κB),in an attempt to explain the pathogenesis of nephropatic syndrome(NS) and the possible mechanisms of Shenkangling.Methods:5.5 mg/kg of Adriamycin was intravenously used to establish rats models with minimal change nephropathy (MCD) and these AN rats were treated with Prednisone in Group C and with Prednisone plus Shenkangling decoction in Group D.Meanwhile, control Groups were established to test changes of NF-B and other pathomorphological indexes in various groups before and 3 weeks after the treatment.Results:There is markedly increased urinary protein,decreased albumin,more active cholesterol,triglyceride and NF-κB activities and aggravated renal injury before the treatment,but all the above-mentioned indexes were significantly improved after the treatment for the AN rats in the treatment group.(P<0.05 or P<0.01)with significantly attenuated renal injury.Conclusion:Animal models created with ADR injection at caudal vein are with similar pathologic manifestations and experimental indexes of “kidney deficiency and blood stasis”in human nephropathtic syndrome,Shenkangling, a TCM decoction with the effect of benefiting the kidney and promoting the blood circulation, can successfully improve such symptoms as kidney deficiency and blood stasis and NF-κB can be used as an objective index for the diagnosis and treatment of the nephrosis of “Kidney Deficiency and Blood Stasis”type in TCM.
