摘 要:目的 探讨SARS恢复期患者特异性IgG抗体滴度水平及变化趋势 ,完善SARS感染后免疫的资料。方法 采用ELISA法检测 2 3例SARS患者发病后 6个月、1 2个月 2个时间点血清中特异性IgG抗体滴度。以单克隆抗体标记 2 3例患者的PBMC ,用流式细胞仪检测HLA A2的阳性率。结果 6个月时IgG抗体滴度的最高值为 1∶1 6 0阳性(4 / 2 3) ,最低值为 1∶1 0阴性 (1 / 2 3) ,平均滴度为 1∶5 7;1 2个月时IgG抗体的最高滴度仅为 1∶80阳性 (6 / 2 3) ,最低值为 1∶1 0阴性 (2 / 2 3) ,平均滴度为 1∶2 7。 2个时间点抗体滴度之间的差异有统计学意义(P =0 0 0 2 )。 2 3例患者中HLA A2阳性 1 4例 ,阳性率 6 0 9%。结论 SARS恢复期患者的抗体滴度在半年以后已呈现明显的下降趋势 ,目前所依据的SARS IgG抗体诊断标准是合理可行的。HLA A2抗原肽疫苗可以治疗和预防 5 0 %左右的SARS病毒感染。Objective To investigate the variation in the titer of SARS specific IgG antibody in convalescent SARS patients, and to establish the immunological data of the patients infected by SARS CoV. Methods The titer of antibodies against the SARS CoV in the sera obtained sixt months and twelve months after onset of the illness from 23 patients recovered from SARS were determined by ELISA assay. HLA A2 alleles of 23 patients were measured through labeling the PBMCs by monoclonal antibody and tested by FACS. Results In these 23 patients, the highest titer in the sixth month was 1∶160(+), the lowest was 1∶10(-), and the average was 1∶57; the highest titer in the twelfth month was 1∶80(+), the lowest was 1∶10(-), and the average was 1∶27. Statistical difference of the titers between the two time points was significant( P =0 002). 14 patients with HLA A2 allele were found in the 23 patients (60 9%). Conclusions The titer of SARS specific IgG antibody in patients recovered from SARS presented a downward tendency after 6 months, so the current diagnostic criteria by determination of SARS IgG isfeasible. HLA A2 restricted peptide vaccine could be used to prevent infections with SARS CoV by about 50%
