白细胞介素-10放免方法的建立及其初步临床应用

摘　　要：本文建立了白细胞介素 - 10 (IL - 10 )放射免疫分析法 (RIA)并初步应用于临床。将重组IL - 10作免疫原 ,获多克隆抗体。采用Iodogen法制备高比活度12 5I -IL - 10 ,建成IL - 10RIA。应用本法测定抑郁症患者组及对照组血清IL - 10含量 ,结果表明 :IL - 10测定范围为 5 - 12 15ng/mL ;对照组血清IL - 10含量为 2 8.0±8.9ng/mL ,而抑郁症患者组IL - 10含量为 2 5 .3± 11.1ng/mL ,较对照组低 (P <0 .0 5 ) ;患者治疗后 (2 8.4± 13.2ng/mL)较治疗前有明显升高 (P <0 .0 5 )。IL - 10RIA的建立为进一步开展IL - 10在相关学科的研究 ,提供了一种有价值的指标。To develop a senstive and specific RIA for interleukin-10(IL-10) and apply it in clinical research, IL-10 was injected into rabbits.A high sensitive and specific antibody against IL-10 was obtained. 125 I-IL-10 was prepared by Iodogen method and purified by Sephadex G-50. The plasma IL-10 level in the healthy volunteers and depression patients before and after treatment was determined by RIA.Results showed that the IL-10 measurement range was 5-1215ng/mL.The plasma IL-10 level of the volunteers was 28.0 ±8.9ng/mL.The IL-10 level(28.4±13.2ng/mL) of depression patients after treatment was significantly higher than that(25.3±11.1ng/mL)before treatment. This developed assay may be a useful technique for related research and clinical application of IL-10.
