玉竹提取物B对Hela细胞凋亡的影响

摘　　要：目的　观察中药玉竹提取物B (EB -PAOA)是否能引起人宫颈癌Hela细胞的凋亡。方法　将体外培养的人宫颈癌Hela细胞与不同浓度的EB -PAOA共育 ,通过倒置显微镜和透射电镜进行形态学观察 ,确认凋亡细胞 ;通过流式细胞仪检测凋亡率和细胞周期的分布。结果　经过EB -PAOA处理后 ,在倒置显微镜和透射电镜下可观察到EB -PAOA引起的典型的凋亡形态学变化 ;流式细胞仪测定显示经EB -PAOA处理后的Hela细胞凋亡率呈时间 -剂量依赖性 ,G0 /G1期细胞减少 ,S期细胞减少 ,G2 /M期细胞增多。结论　EB -PAOA能够诱导人宫颈癌Hela细胞凋亡 ,EBObjective To observe whether the extract B of Polygonatum Odoratum (EB-PAOA) can induce the apoptosis in human cervix cancer Hela cells.Methods Human cervix cancer Hela cells were cultured in vitro with different concentration of EB-PAOA. Cells were observed and confirmed whether there were apoptotic cells with an inverted microscope and electronmicrograph. Apoptotic rate and cell cycles were then detected through a flow cytometer.Results It can be observed that large number of apoptotic cells had representative morphological characteristics under the inverted microscope and electronmicrograph. Though the detection of the flow cytometer, it can be seen that the apoptotic rate of cells was of time and dose dependence. The analysis result of cell cycle indicated that the number of cells of G 0/G 1 and S period was reduced, and the number of G 2/M period was increased.Conclusions EB-PAOA can induce the apoptosis of Hela cells, which may be one of the mechanism of EB-PAOA's inhibition effect on Hela cells.
