Research on Formation of Plant from a Piece of Lower-Plumular Axis of the Aseptic Seedling of Cauliflower in Vitro Culture

摘  要:花椰菜无菌苗下胚轴切段。接种于MS培养基附加KTO.5mg/L及NAAO.5mg/L诱导生根生芽,经一次培养35~40天形成完整植林。然后接种于MS+KTO.1mg/L+NAAO.1mg/L培养基进行继代培养25天100%形成完整植株。由于生出的芽与根由下胚轴直接连结,因此植株成活率高,成苗速度快,同时取材也不受大田和季节的限制。从而为保持优良品种种质及杂一代的快速繁殖提供了可靠途径。花椰菜试管苗往外移栽容易成活,一周内注意保温保湿90%以上可成活,因而为组织培养苗应用于生产提供了有利条件。试管苗经两年栽到地里观察“3198”及瑞士雪球两品种结球正常,因此花椰菜下胚轴切段离体培养对花椰菜种性的提纯,良种及杂一代的快速繁殖有一定意义。A Piece of lower-plumular axis of aseptic seedling of Cauliflower was inoculated on MS media in which 0.5mg/1 kT and 0.5mg/1 NAA were added, in order to induce rooting and sprouting. After 35——40 days, the intact plant was formed. The Plant was inoculated again on MS media with 0.1mg/1 KT and 0.1mg/1 NAA as successive culture,After 25 days,intact plant was formed with survival rate of 100 per cent.The survival rate of plant was high,and the speed of young plant establishment was fast, dewing to the bud and root produced were touched directly with lower-plumular axis,and that of material collecting were not restricted in seasons and field condition.Thus, a reliable way for maintaining good quality of varieties and reproducing F_1-hybrids quickly were provided Test-tube seedlings of cauliflower which were transplanted could survive easily with survival rate over 90 percent,if a suitable temperature and moisture within one week.So,this provided a good conditon for tissue culture seedlings to be applied in production Having grown for two years, test-tube seedlings of both varieties '3198' and 'Swiss—snow ball' could bear normally after they were trans planted. Therefore, it is of significance for a piece of lower plumular axis of Cauliflower in vitro cultre to be used to purify seeds, to reproduce a good quality of seeds and F_1-hybrids quickly.
