摘 要:目的 建立稳定的可重复的大鼠脑局部低灌注模型 ,并通过CT灌注等方法验证其可靠性。方法 2 7只实验用Wistar雄性大鼠随机分为三组 (假手术对照组、低灌注 6h组和脑梗死 6h组 )。模型的制备采用改良的大脑中动脉线栓法 ,在术中应用激光多普勒血流仪实时监测局部大脑中动脉区脑血流量 ,术后利用CT灌注成像对血流状况进行观察 ,并与病理检查、TTC染色等对照。结果 梗死组CT灌注参数rCBF较低灌注组明显下降 ,病理检查呈典型脑梗死改变 ;低灌注组仅见脑缺血病变。结论 在线栓法制备脑缺血模型的基础上 ,应用激光多普勒血流仪实时监测术中血流状态 。ObjectiveTo establish a stable and reliable model of local cerebral blood hypo perfusion in rats, and to evaluate it by CT perfusion and histopathological study. MethodsTwenty seven male Wistar rats were divided into three groups, nine rats in each group (the control group, hypo perfusion group and infarction group). A nylon thread was inserted into the right middle cerebral artery (MCA), and the same time the region cerebral blood flow (rCBF) was examined by the laser Doppler blood perfusion monitor. About 6 hours after operation, all the rats were examined by CT perfusion, and 3 of each group were performed by TTC stain and histopathological study. ResultsThe rCBF of both infarct group and hypo perfusion group were reduced on CT perfusion, and rCBF of the infarct group was even lower than that of hypo perfusion group. In histopathological results, the brain in hypo perfusion group showed astrocytic swelling. Most of the neuronal was normal, but in the brain of infarct group, severe neuronal degeneration and necrosis could be found. ConclusionBased on the thread method, using laser Doppler blood perfusion monitor to monitor the blood flow during operation, we can establish stable and reliable model of local cerebral blood hypo perfusion in rats.
