摘  要:目的 FTY720和40-氧-(2-羟乙基)-雷帕霉素(RAD)是两种新型免疫抑制剂。为评估这两种药物的效果,我们以小鼠心脏移植为模型,对比观察了这两种药物和环孢素A(CsA)对移植物存活时间的影响。方法 供体为BALB/C小鼠,受体为C57BL/6小鼠,受鼠随机分为4组,每组6只:A组空白对照;B组每日管饲CsA 10 mg/kg体重;C组每日管饲RAD 3 mg/kg体重;D组每日管饲FTY 3 mg/kg体重。各服药组均由移植当日开始喂药,至移植心停跳或术后14 d停药。结果 各组的存活天数分别为:A组6、7、8、8、9、9;B组9、9、10、11、12;C组10、12、13、13、14;D组10、12、16、16、17、18。B、C组各有1例带心死亡。所有移植心都出现程度不同的急性排斥反应征象。结论 单独使用RAD或FTY可显著延长小鼠心脏移植物的存活时间。在各所选剂量上,CsA和RAD差异无显著性(P>0.05),FTY的效果明显优于CsA和RAD。FTY和RAD是较有前途的新型免疫抑制剂。Objective To campatively study the effect of FTY720 and 40-O-(2-hydroxyethyl) rapamycin (RAD) (two kinds of new synthetic immunosuppressant) and cyclosporine A (CsA) on the survival of mouse heart graft by using mouse heart graft as a model. Methods BALB/c to C57BL/6 mouse cervical heterotopic heart transplantation was used as a model. The recipient mice were randomly divided into group A (blank control group), group B (CsA group, 10 mg/kg body weight every day), group C (RAD, 3 mg/kg body weight every day) and group C (FTY720, 3 mg/kg body weight every day) respectively by oral gavage, n = 6 in each group. The treatment started at the day-0 and stopped at the day-14 or when the heart graft stopped beating. Results The survival time (days) of the grafts in groups A was 6, 7, 8, 8, 9, 9 respectively;in group B 9, 9, 10, 11, 12 respectively;in group C, 10, 12, 13, 13, 14 respectively; in group 4 10, 12, 16, 16, 17, 18 respectively. There was one mouse which died with functional graft in group-B and group 3 respectively. All grafts were subjected to acute rejection reaction with different grades. Conclusion FTY720 or RAD could significantly prolong the survival time of mouse allograft. There was no significant difference between RAD and CsA, while FTY720 had better effects than RAD and CsA significantly.
