摘  要:目的 观察转染人转化生长因子 β3 (hTGF β3 )基因对瘢痕成纤维细胞Ⅰ、Ⅲ型胶原合成的影响。方法 通过脂质体介导的方法 ,将人表皮细胞生长因子基因真核表达质粒 (pBK CMV) TGF β3 质粒转染体外培养的增生性瘢痕成纤维细胞 ,采用RT PCR法检测TGF β3 mRNA的表达 ,放射免疫 (RIA)法测定细胞培养上清液中Ⅰ型和Ⅲ型前胶原的浓度。结果 构建的TGF β3 质粒可成功转染成纤维细胞 ,并增强Ⅰ型和Ⅲ型胶原的表达 ,下调Ⅰ /Ⅲ型胶原比值。结论 转染hTGF β3Objective To observe the effects of transforming factor β 3(TGF-β 3) gene transfer on fibroblasts from hypertrophic scar and the type Ⅰand Ⅲ collagen synthesis.Methods Fibroblasts were harvested from hypertrophic scar and then were transferred with pBK-CMV-TGF-β 3 plasmid mediated by Lipofectamine. The TGF-β 3 mRNA expression was assayed by RT-PCR. The type Ⅰand Ⅲ collagen contents in the culture supernatant were determined by RIA.Results pBK-CMV-TGF-β 3 plasmid could be transfected into the fibroblasts successfully. The gene-transfected fibroblasts increased the secretion of type Ⅰand Ⅲ collagen and downregulated the ratio of typeⅠand Ⅲ collagen.Conclusion Human transforming factor β 3(hTGF-β 3) gene can regulate the biological activity of fibroblasts from hypertrophic scar.
