摘 要:目 的 观 察 5-氟 尿 嘧 啶 (5-FU )对 体 外 培 养 不 同 兔 龄 晶 状 体 上 皮 细 胞 (RLEC)的 作 用 。 方 法取 体 外培 养 第 4 代 的两 组 RLEC 接种 于 24 孔 培 养 板:①继 续 培 养 48 h 后 ,加 入 不 同 浓 度 的 5-FU (0.2、0.4、0.8、1.6、3.2 μg/m l)作用 24、48 和 72 h 后 ,做 活 细 胞 计 数 ,求 出 半 效 抑 制 量 (ID50);②细 胞 传 代 后 立 即 加 入 5-FU 24 hID50 浓度 的 1/10,作 用 24 h 后 ,计算 细 胞 贴 壁率 ;③培养 24 h 后 加 入 不 同 浓 度 的 5-FU ,作 用 24 h 后 掺 入 氚 标 记胸 苷 (3H -TDR ),16 h 后 做 液 体 闪 烁 测 量 计 数 。 结 果 5-FU 作 用 幼 年 组 RLEC 24、48 和 72 h 的 ID 分 别 为 500.88、0.74 和 0.65 μg/m l;作 用 成 年 组 R LEC 24、48 和 72 h 的 ID50 分别 为 0.86、0.61 和 0.37 μg/m l。 5-FU 作 用 成年 组 R LEC 细胞 贴壁 率 明显 低于 幼 年组 (P<0.05?Objective To study the effect of 5-fluorouracil (5-FU) on the proliferation of different-aged rabbit lens epithelial cells (RLEC) in vitro. Methods The young and the adult RLECs were cultured in vitro. The fourth passage of the two groups RLECs was used for assay and inoculated into 24-well culture plates. ① After a 48-hour incubation, treated with various concentrations of 5-FU (0.2, 0.4, 0.8, 1.6, 3.2 μg/ml) for 24, 48 and 72 hours, the ID50 values was determined by counting the RLEC numbers on a counting plate; ② After a passage culture, one tenth of 24 hours′ ID50 was added into the culture medium immediately, and cultured for 24 hours to investigate the rate of attached cells; ③ Aftera 24-hour incubation, treated with various concentration of 5-FU for 24 hours, 3H-thymidine was added; after 16 hours, the radioactivity was determined by liquid scintillation counting. Results The ID50 of 5-FU exposed to the young RLECs for 24, 48 and 72 hours was 0.88, 0.74 and 0.65 μg/ml respectively; and that to the adult RLECs for 24, 48 and 72 hours was 0.86, 0.61 and 0.37 μg/ml respectively. In the adult group, the rate of attached cells was less than that in the young group (P<0.05). 5-FU inhibited cellular DNA synthesis effectively. In the adult group, the inhibition was stronger than in the young group. Conclusion 5-FU has a significant inhibition effect on different-aged RLECs. The effect is in a dose-dependent and time-dependent manner. The adult RLECs are easier to be inhibited than the young group by 5-FU.
