绝经期后女性腹部脂肪组织芳香化酶mRNA表达的研究

摘　　要：目的 :研究绝经后妇女腹部皮下、大网膜脂肪组织中芳香化酶mRNA的表达水平变化。方法 :应用半定量逆转录 -聚合酶链反应 ,检测绝经后肥胖妇女及非肥胖正常妇女芳香化酶mRNA的表达 ,分析其与空腹真胰岛素水平、血糖、血脂、体质量指数、腰臀比及血压等指标的关系。结果 :绝经后肥胖妇女皮下、大网膜脂肪组织芳香化酶mRNA的表达水平高于正常体质量的绝经后妇女。绝经后女性皮下脂肪组织芳香化酶mRNA的表达水平高于大网膜脂肪组织。绝经后肥胖女性皮下、大网膜脂肪组织芳香化酶mRNA的表达与腰臀比、空腹胰岛素呈正相关。结论 :绝经后女性腹型肥胖及与肥胖相关的胰岛素抵抗与腹部脂肪组织内芳香化酶mRNA表达水平有关。Objective:To study the aromatase mRNA expression in subcutaneous(SC)and omental(OM)adipose tissue of postmenopausal women.Methods:P450AROM mRNA was measured by a semi-quantitative assay based on reverse transcribing the mRNA into cDNA with reverse transcriptase(RT)then amplification of the cDNA using the polymerase chain reaction(PCR).To analyze the relation with fasting plasma glucose and fasting plasma insulin,total cholesterol,serum triglyceride,body mass index,waist to hip ratio,and blood pressure.Results:In postmenopausal women,the aromatase mRNA expression in SC was significantly higher than that in OM adipose tissue.The level of aromatase mRNA in obesity group was significantly higher than that in control at both sites.Aromatase mRNA expression in SC and OM adipose tissue in the obesity were positively correlated with waist to hip ratio and fasting plasma insulin.Conclusion:In postmenopausal women,central obesity and obesity related with insulin resistance contributed to the expression of aromatase mRNA in abdominal adipose tissue.
