摘  要:目的:比较姜黄醇提取物中不同组分对常规培养血管平滑肌细胞(Vascular Smooth Muscle CellsVSMC)增殖的影响和对血管紧张素Ⅱ(AngⅡ)条件培养的平滑肌细胞增殖作用的影响。方法:姜黄醇提取物经制备型薄层色谱分离,纯化,含量测定后配制成不同浓度的药物溶液,分别与常规培养的VSMC和浓度为2μmol/L的AngⅡ条件培养VSMC中共孵育48h,MTT法测定细胞的增殖情况。结果:姜黄素、一脱甲氧基姜黄素和二脱甲氧基姜黄素在剂量为(1.35×10-5mol/L)时,对常规条件培养和浓度为2×10-6mol/L的AngⅡ条件培养的VSMC均有明显的抑制增殖作用,P<0.001-0.05;在剂量为(5.4×10-7mol/L)时三者均无明显差异,存在剂量依赖性。抑制作用姜黄素>一脱甲氧基姜黄素>二脱甲氧基姜黄素;对.AngⅡ条件培养的抑制率>常规条件培养。色谱的原点物质给予高剂量姜黄素组相同剂量,无抑制作用。Objective: Compare the effect of different component abstracted from Curcumin on proliferation of Vascular Smooth Muscle Cells (VSMC) cultured and normal condition with proliferation of VSMC induced by Ang Ⅱ. Methods: The component abstracted from Curcumin was made into various concentration liquid after TLC, puring, measuring. The VSMC proliferation was measured respectively after the two sort VSMC has been cultured for 48h with the liquid. Results: When the liquid is 1.35 × 10 -5 mol/L, Curcumin, Demethoxy Curcumin and Bisdemethoxy Crucumin respectively have significant inhibition on VSMC proliferation. When it is 5.4 ×10-7mol/L, they have no significant differences. The inhibition depended on dose. The inhibition of curcumin is stronger than Demethoxy Curcumin and Bisdemethoxy Curcumin. Curcmin can strongly inhibit the proliferation of VSMC induced by Ang Ⅱ .
