复方茯苓甘草汤对低氧大鼠磷脂酶A2活性及其炎症介质的影响

摘  要:低氧性肺动脉高压(hypoxic pulmonary hypertension,HPH)是慢性阻塞性肺疾病(chronic obstruc-five pulmonary disease,COPD)发展过程中的特征性表现之一,实验通过建立HPH大鼠动物模型,研究复方茯苓甘草汤对磷脂酶A2(phospholipase A2,PLA2)及其炎症介质血栓素B2(thromboxane B2,TXB2)、6-酮-前列腺素F1α(6-keto-prostaglandin F1α,6-keto-PGF1α)的影响,以探讨其作用机制。目的:从磷脂酶A2(PLA2)活性及其相关炎症介质途径研究复方茯苓甘草汤防治常压慢性低氧性肺动脉高压的作用机理。方法:30只清洁级健康雄性SD大鼠随机分为正常对照组、单纯缺氧组和低氧加复方茯苓甘草汤治疗组。右心导管法测定大鼠肺动脉压(mPAP),计算右心肥大指数,微量滴定法检测血浆、肺匀浆和支气管肺泡灌洗液(BALF)中PLA2活性,放射免疫分析法检测血浆及BALF中血栓素B2(TXB2)、6-酮-前列腺素(6-keto-PGF1α)活性。结果:连续低氧3周后大鼠mPAP、右心肥大指数、血浆、肺匀浆及BALF中PLA2活性、TXB2、6-keto-PGF1α含量显著增加,6-keto-PGF1α活性下降。复方茯苓甘草汤可有效逆转上述改变。结论:复方茯苓甘草汤可通过抑制PLA2及其炎症介质途径起到防治低氧性肺动脉高压的作用。Background & Objective: Chronic obstructive pulmonary disease (COPD) is a common medical problem, Which constitutes a major health burden worldwide. In patients with COPD, pulmonary hypertension is associated with a worse prognosis. The present investigation was focused on the role of phospholipase A2, and its related inflammatory mediators, thromboxane B2(TXB2 ) , 6 - keto - prostaglandin F1α(6 - keto - PGF1α) in HPH and the effects of Fufang Fulin Gancao Decoction. Methods :30 healthy Spraque - Dawley male rats of three months old weighted 260 ± 20g were randomly divided into three groups : normal control group(group C, n = 10, perfused normal saline 5ml ·d -1); chronic hypoxic group ( group H, n = 10, perfused normal saline 5ml · d-1) ; hypoxia plus Fufang Fulin Gancao Decoction treatment group (group HF, n = 10, perfused Fufang Fulin Gancao Decoction 5ml · d-1 before hypoxia). We established the rat model of chronic hypoxic pulmonary hypertension by exposing rats to normobaric hypoxic conditions for 3 weeks, and rats of group C were used as normoxic controls. Mean pulmonary arterial pressure (mPAP) was measured by a right cardiac catheterization. Right ventricular hypertrophy was expressed as the ratioes of right ventricular wall to left ventricle plus septum[RV · ( LV + S)-1]. The PLA2 activity, TXB2, 6 -keto-PGF1α in bronchial alveolus lavage fluid (BALF) , pulmonary plasma and pulmonary homogenate were measured by modified microtitrimetic methods and radioimmunoassay, the data were analyzed for statistical difference using one -way analysis of variance (ANOVA) of the statistical package for social science (SPSS, 10.0 version). Significance levels was established with a P value less than 0.05. Results: Compared with the control group, hypoxic rats developed remarkable pulmonary hypertension and increment in the right ventricular hypertrophy ( P < 0.01).
