补肾活血及益气活血方药对脑缺血老龄大鼠Ca^(2+)和Ca^(2+)-ATPase的影响

摘　　要：目的探讨缺血性脑损伤的病理,并比较补肾活血与益气活血方药对其防治作用,为临床运用提供依据。方法复制老龄大鼠急性脑缺血模型,观察益元活血丹及益气活血方对脑缺血老龄大鼠脑组织病变、Ca2+-ATPase活性和Ca2+含量的影响。结果与假手术组比较,模型组脑组织病理损伤明显,Ca2+含量明显增加,Ca2+-ATPase活性显著下降;与模型组比较,各用药组脑组织病理损伤明显减轻,Ca2+含量均明显降低,益气活血组脑组织Ca2+-ATPase活性增加。结论老龄大鼠急性脑缺血损伤与脑组织Ca2+-ATPase活性下降及Ca2+含量升高有关;益元活血丹及益气活血方对急性脑缺血防护作用显著,其作用机制有所侧重:相同之处在于均能降低Ca2+含量;不同之处在于益气活血方药提高Ca2+-ATPase活性更为明显。To provide the patholigy of ischemic brain tissue injury and the mechanism of the two combination of TCM Decoction,and experimental comparison of them and a basis for the clinical application in the field of ICVD of the elderly.Methods:Via the model of both common carotid arteries(CCA) obstructed. The study is purposely to observe the effect of the Tonifying the Kidney and Activating Blood Circulation Decoction,Yi Yuanhuoxuedan (益元活血丹,YYHXD),and the Tonifying Qi and Activating Blood Circulation Decoction,Yiqihuoxuefang(益气活血方,YQHXF).To assay the pathological anatomy,the changes of Ca2+-ATPase activity,Ca2+content in the brain tissue,too.Results:Firstly,compared with the pesudo-surgery group,the damage of the brain tissue was great.The brain tissue Ca2+content rised significantly,while Ca2+-ATPase activity declined. Secondly,contrasted with the eldly model group,the ischemic damage was great lower,and so did the Ca2+Content. Ca2+-ATPase activity of YQHXF group rised.Conclusion:The ischemic brain injuries in aged rats were clear and possibly relative to the weakening of Ca2+-ATPase activity and calcium overload.It showed the YYHXD and YQHXF could significantly reduce the cerebral injuries caused by ischemia.Hence, the functional mechanism might concern with mitigating the Ca2+content of the brain tissue.The prevence of the TCM herbs both rested in depressing of Ca2+content.The YQHXF herbs raised Ca2+-ATPase activity more significantly than the other.
