外源性一氧化氮抑制剂和诱导剂对高血压大鼠主动脉平滑肌细胞增殖的影响

摘  要:目的:观察长期使用Nω-硝基-L-精氨酸(Nω-nitro-L-Arginine,L-NNA)和谷氨酸单氨钠(L-Glutamicacidmonoammoniumsalt,MSG)后高血压大鼠主动脉中膜的变化。方法:将60只SD大鼠随机分为4组,一组作为对照组,一组用双肾双夹法建立高血压大鼠模型,另2组首先建立高血压模型,分别腹腔内注射L-NNA犤0.2mg/(100g·d)犦和MSG犤50mg/(100g·d)犦,用测微尺测量主动脉中膜的厚度及平滑肌细胞的层数,并计算中膜的面积。结果:中膜平滑肌细胞层数和面积的变化具有相似性,高血压组中膜平滑肌细胞层数和面积虽然7d变化不显著,但随着高血压持续时间的延长30,70d均明显增多(t=3.257,3.986,3.784,4.251,P<0.05),70d分别达(28.3±0.8)层和(1031.4±39.7)×103μm2;L-NNA组高血压大鼠30,70d与高血压组大鼠比较显著增多(t=3.087,3.469,3.895,3.884,4.011,P<0.05),且随着高血压的发展有增多趋势,其中70d为(32.1±0.5)和(1159.5±46.7)×103μm2;应用MSG组大鼠7,30d较L-NNA组大鼠明显减少(t=4.779,4.989,3.683,4.709,P<0.05),与高血压组无明显差异,在70d减少明显(t=3.784,P<0.05),为(22.3±0.5)和(826.3±36.4)×103μm2。结论:抑制一氧化氮生成可升高血压,并促进血管平滑肌细胞增殖,有效促进一氧化氮生成可减少血管中膜平滑肌细胞?AIM:To investigate the changes of aortic tunica media in hypertensive rats after long term application of Nω nitro L arginine(L NNA) and L glutamic acidmonoammonium salt(MSG). METHODS: Totally 60 rats were randomly divided into 4 groups: control group, hypertensive group(hypertensive models were established with the method of clamping bilateral renal arteries of rats), hypertensive models were established at first in the other two groups, and then were administered by intraperitoneal injection of L NNA (0.2 mg/100 g per day)and MSG(50 mg/100 g per day) respectively. The thickness of aortic tunica media and layers of smooth muscle cells(SMCs) of the hypertensive rats were measured with micrometer to calculate the area of tunica media.RESULTS:The changes of SMCs layer of tunica media were similar to that of area.In the hypertensive group, there were no significant changes in the layers and area of SMCs in tunica media at 7 days,but with the elongation of hypertension,the area and thickness of SMCs were increased markedly at 30 and 70 days(t=3.257,3.986,3.784,4.251,P< 0.05),which were(28.3±0.8) layers and(1031.4±39.7)×103 μm2 at 70 days; Those in the L NNA groups were significantly increased at 30 and 70 days as compared with those in the hypertensive group(t=3.087,3.469,3.895,3.884,4.011,P< 0.05),and had an increasing tendency with the development of hypertension,which were(32.1±0.5) layers and(1 159.5±46.7)×103 μm2 at 70 days;Those at 7 and 30 days in the MSG group were obviously decreased as compared with those in the L NNA groups(t=4.779,4.989,3.683,4.709,P< 0.05),but insignificantly different from those in the hypertensive group, and obviously decreased at 70 days[(22.3±0.5) layers and(826.3±36.4)×103 μm2](t=3.784,P< 0.05).CONCLUSION: Inhibition of NO production can increase blood pressure and accelerate the proliferation of SMCs. Effective acceleration of NO production can reduce the proliferation of SMCs in tunica media, and its mechanism may be related with the inhibitive effect of NO o
