大鼠同种肝移植排斥反应中重组人IL-10对TNF-α及IL-2的影响

摘　　要：目的　探讨在大鼠同种肝移植排斥反应中重组人白细胞介素 10 (rhIL 10 )对肿瘤坏死因子 α(TNF α)及白细胞介素 2 (IL 2 )的影响。方法　供体为ACI(RTIα)大鼠 ,受体为LEW (RTI1 )大鼠。实验组大鼠在移植前后腹腔注射 10 μg kg·drhIL 10。对照组腹腔内注射 0 .85 %等量生理盐水 ,注射时间同实验组。用免疫组化方法观察肝脏组织中TNF α及IL 2的改变。结果　实验组大鼠平均存活时间 (13.0± 1.0 )d ,与对照组 (9.4± 0 .7)d相比 ,有非常显著性意义 (P <0 .0 0 1) ;移植后各时间点实验组比对照组肝组织破坏程度减轻 ,TNF α阳性率明显小于对照组 (P <0 .0 1) ,IL 2阳性率显著小于对照组 (P <0 .0 0 1)。结论　rhIL 10能够降低大鼠移植肝组织中IL 2和TNF α的表达 ,减轻排斥反应 ,延长移植物的存活时间 ,表明rhIL 10对肝移植有积极的作用。Objective To investigate the role of recombinant human interleukin-10(rhIL-10) in acute rejection in rats after orthotopic liver homeo transplantation.Methods ACI(RTIα) rats and LEW(TRI1) rats were used as donors and receptors,respectively.Treated group received injections of rhIL-10 at dose of 10μg/kg·d pre- and post-transplantation.Control group received the same dose of 0.85% sodium chloride,the injection time was the same with the treated group and the change of IL-2 and TNF-α in the liver tissue was measured with immunohistochemical method.Results The mean survival time of treated group(13.0±1.0)d was much longer than that of control group(9.4±0.7)d(P<0.001).The average concentration of IL-2(P<0.001) and TNF-α(P<0.01) in the hepatic tissue of treated group decreased obviously compared with the control group.The grade of liver injures in treated group after transplantation was less than that of control group.Conclusion rhIL-10 can decrease expression of IL-2 and TNF-α,relieve the severity of acute rejection after liver transplantation and prolong the survival time of grafts.And these results show that rhIL-10 therapy is useful in liver transplantation.
