摘  要:目的:观察阿妥伐他汀对高糖培养的大鼠肾小球系膜细胞增生的作用,并探讨其作用机制。方法:用不同浓度的阿妥伐他汀刺激高糖培养的系膜细胞,用4-甲基偶氮四唑蓝(tetrazoliumsalt,MTT)法观察其增殖情况,并通过流式细胞仪检测其细胞周期变化。结果:阿妥伐他汀在一定范围内以剂量依赖及时间依赖关系抑制高糖诱导的肾小球系膜细胞过度增生,并使G0鄄G1期细胞比例增高,S期细胞数减少,促进过度增殖的系膜细胞凋亡。结论:阿妥伐他汀能够抑制高糖诱导的系膜细胞增生,这种作用可能是通过抑制细胞分裂、复制以及促进细胞凋亡实现的。Objective: To investigate the effect of atorvastatin on proliferation and cell cycle of rat mesangial cells treated with high concentration of glucose. Methods: Mesanngial cells were incubated with different concentrations of Atorvastatin under high glucose condition. Cell proliferation was assessed by MTT colorimetric assay. Cell cycle was detected by flow cytometry. Results: Ranging from 0.1 to 40.0 μmmol/L, atovastatin can inhibit the proliferation of mesangial cells under high glucose concentration. Atorvastatin also increases the apoptosis of mesangial cells under high glucose condition. Conclusion: Atovastatin can inhibit cell division and replication and promote apoptosis to exert inhibitory effect on the proliferation of mesangial cells under high glucose concentration.
