摘  要:作者建立了大鼠睾丸细胞膜的绒毛膜促性腺激素受体的放射性配体结合分析。睾丸经匀浆、蔗糖密度梯度离心及高速离心制成膜制剂。^(125)I-HCG与膜制剂于30℃反应1小时半,测得膜受体容量为0.36×10^(-12)mol/μg蛋白,K_D值为0.45×10^(-10)mol/L,Hill系数为0.976,k_1为0.41×10^(-3)pmol/min,k_2为2.12×10^(-3)pmol/min。解离图不成直线,分快相和慢相。A radioligand-receptor bind-ing assay for human chorionic gonadotro-pin (HCG) has been established with cellmembranes preparation of rat testis.Par-tially purified cell membranes preparationwas prepared from rat testis by sucrosedensity gradient ultracentrifuge.The bind-ing assay was performed by incubating^(125)I-HCG with rat testis cell membranespreparation at 30℃ for 1.5h in a totalincubation volume of 0.5ml. Scatchardplot analysis for the binding assay revealeda linear binding plot and yielded a boundcapacity of testis cell membranes receptorsof 0.36×10^(12)mol/μg protein. K_D of 0.45×10^(-10)mol/L. Hill coefficient was 0.976.Studies on the kinetics of the reaction ofradioligand-receptor binding showed thatthe mean values for the association rate(k_1) and dissociation constants (k_2) wereindividually 0.41×10^(-3)pmol/min and 2.12×10^(-3)pmol/min; the dissociation plot wastwo-phase, with 15×10^(-3)pmol/min offast-phase and 1.5×10^(-3)pmol/min of slow-phase.
