Nilestriol Prevents Osteoporosis in Ovariectomized Rats

摘  要:作者选用4月龄雌性SD大鼠随机分为:卵巢切除组,假手术组(开腹后关腹)及切除卵巢术后给予尼尔雌醇(CEE_3)组。按每100g体重给CEE_3 0.15mg灌胃,每周1次,共10周。取右侧胫骨制成不脱钙骨切片进行骨计量学测定,并测定肱骨重量,无机质重量及骨钙、磷含量等。结果:从卵巢切除组骨小梁体积,骨小梁宽度、骨重量和无机质重量与体重之比及骨钙含量等方面显示绝经后骨质疏松动物模型建立。而尼尔雌醇组各指标与假手术组相似(P>0.05),表明尼尔雌醇通过抑制过高的骨转换,恢复骨再建过程的平衡而防止切除卵巢大鼠的失骨,提示可用于预防绝经后骨质疏松。In order to determine theeffects and mechanism of Nilestriol uponbone metabolism, 4-month aged femaleSD rats were divided randomly into threegroups: The OVX group subjected to bila-teral ovariectomy, the SHAM group tosham operation, the Nilestriol (CEE_3)group to bilateral ovariectomy and treatedwith CEE_3 0.15mg/100g BW once a weekfor 10 weeks. The CEE_3 was introduceddirectly into the stomach from the firstday after operation. All rats were killed10 weeks after operation and their rightproximal tibiae were processed undercal-cified for quantitative bone histomorpho-metry. The weights of bone and inor-ganic matter(ash content) and content ofcalcium, phosphorus of right humeralwere measured. The results showed thatin the OVX group the trabecular bonevolume, the trabecular thickness,the con-tent of calcium bone, and the ratios ofbone weight and ash weight to bodyweight wete all significantly reduced.Thebone loss was associated with an increasein bone remodeling; the bone resorptionappeared much more marked than the boneformation. In contrast, the ovariectomizedanimals treated with CEE_3 had normaliza-tion of bone mass and a declined indexof bone resorption and formation whichrecovered the balance of bone remodeling.It suggested that CEE_3 is successful inprevention of bone loss in OVX rats. It isconvinced of using Nilestriol to preventpostmenopausal osteoporosis.
