摘  要:利用AFLP技术从京玉、美味正反交F1代69株群体中筛选到3个特异扩增片段,与桃果实非酸/酸性状紧密连锁。回收特异片段并将其克隆、测序,结果表明特异片段A、B、C均来自于EcoRI与MseI两内切酶酶切片段,大小依次为140、199、408bp,利用序列分析软件推测片段B、C中可能含有与糖水平调控作用相关的因子。上述结果对AFLP标记向SCAR标记的转换具有重要意义。Three specific amplified DNA fragments linked to fruit non-acid /acid trait of peach were obtained from 69 F1 seedlings of the intra-specific cross between cultivars 'Jingyu' and 'Flavortop' by using AFLP technique. After cloning the specific fragments and sequencing,we found that fragment A,B and C all were digested by both EcoRⅠand MseⅠ,and were 140 bp,199 bp,and 408 bp in length respectively. It was presumed that fragments B and C might contain elements which are relevant to regulating the sugar content by utilizing sequence analysis software. This study would be important for further SCAR marker conversion.
