摘 要:利用RAPD技术对国内外26份西瓜材料进行遗传亲缘关系研究、构建每份材料的特异指纹。在720个RAPD随机引物中筛选出15个引物用于聚类分析,将供试26份材料分为6个类群1个美国生态型类群、2个中间生态型类群、1个东亚生态型类群、2个野生类群。将京欣一号杂种纯度鉴定的RAPD特异标记K14-1500和N04-600成功转化为SCAR标记,并应用于京欣一号纯度鉴定实践。RAPD analysis was applied to assess the genetic relationships among 26 accessions of watermelon [Citrullus lanatus(Thunb.) Mansf.], and to construct fingerprinting of these accessions. Results of clustering analysis based on polymorphism amplified with the fifteen primers which were screened from 720 arbitrary 10-mer primers showed that the 26 accessions were assigned to 6 groups which were 1 East-Asia group, 1 American group, 2 medial groups and 2 African wild groups. RAPD makers, K14-1 500 bp and N04-600 bp were successfully transformed to SCAR markers, and were used for identification of the purity of watermelon hybrid Jingxin №.1.
