摘  要:目的:采用复合造模法建立绝经期糖尿病骨质疏松症大鼠模型,从生物力学角度探讨双黄益骨方的防治作用、量效关系及作用机制。方法:将210只6个月龄雌性清洁级SD大鼠随机分为假手术组、模型组、预防组和治疗组(包括高剂量中药、低剂量中药组)并与尼尔雌醇组、立普妥组对照。除假手术组外,均行卵巢摘除术及注射链脲佐菌素诱导绝经期糖尿病骨质疏松症。分别于术后第10,24周取各组大鼠左股骨和L4行最大弯曲力、最大压缩力和最大凹入应力检测。~5结果:第24周假手术组的最大压缩力307.905±51.057)N犤(和最大凹入应力犤(70.180±15.269)N犦显著大于第10周假手术组犤(234.064±35.390),(50.986±9.925)N犦(P<0.05),而模型组第24周与第10周比较则呈下降趋势;模型组、中药预防组最大凹入应力较假手术组明显减低(P<0.05);第24周模型组最大压缩力、最大凹入应力分别为(犤137.014±72.560),(22.096±17.350)N较假手术组显著犦降低(P<0.001);高剂量中药组与尼尔雌醇组较第24周模型组最大压缩力显著提高犤分别为(212.698±54.661),(256.118±85.429)NP<0.05~0.01)。低剂量中药组与立普妥组在改善骨力学方面犦(未能显示出显著疗效。结论:正常组骨强度和变形力随月龄增长而提高,模型组则呈下降趋势,成模后,模型组较正常?AIM:To study the preventing effect,dose effectiveness relationship and action mechanism of shuanghuang yigu recipe from the respect of biomechanics in postmenopausal osteoporosis diabetic rats model made with compound animal mode making method. METHODS:Totally 210 female SD rats aged 6 months were randomly divided into sham operate group,model group,preventive group and treatment group(including the high dose decoction group and the low dose decoction group),and compared with the nilestriol group and the lipitor group.Except for the sham operate group,all the animal models were performed with ovariectomy and injected streptozotocin to induce postmenopausal osteoporosis diabetes.The stress examination of maximum bending force(MBF),maximum compressing force(MCF) and maximum depressing force(MDF) were performed to left femur and L4-5 at the 10th and 24th weeks after operation respectively. RESULTS:MCF[(307.905±51.057) N] and MDF[(70.180±15.269) N]at the 24th week after operation were remarkably greater than those at the 10th week after operation in the sham operate group[(234.064±35.390),(50.986±9.925) N](P< 0.05),but decreased in the model group.MDF in the model group and the decoction preventive group significantly decreased compared with the sham operate group(P< 0.05).MCF and MDF in the model group at the 24th week after operation significantly decreased compared with the sham operate group[(137.014±72.560), (22.096±17.350) N respectively](P< 0.001);Compared with the nilestriol group, MCF significantly increased in the high dose decoction group[(212.698±54.661), (256.118±85.429) N respectively](P< 0.05 to 0.01).There was no significant effect on improving bone mechanics in the low dose decoction group and the liptor group. CONCLUSION:The stress and bone intensity in normal group increase as months pass by, and decrease in the model group in the contrary.When the models are done, the significant decrease in bone intensity is found in the model group compared with the normal group that indicates s
