摘 要:目的:探讨用三种不同的方法测定嗜酸性粒细胞的数量,以了解不同方法间的准确性和相关性.方法:①采用全血单克隆抗体CD16荧光标记方法,用流式细胞仪检测细胞CD16-粒细胞的百分率.②应用SYSMEX SE-9500全自动血细胞分析仪进行检测.③用显微镜直接计数.结果:三种方法所测定的结果没有统计学差异(P>0.05),并且方法间具极好的相关性(r=0.99).结论:显微镜直接计数方法操作较烦琐,重复性稍差.五分类血细胞自动分析仪计数方法操作简便,重复性好.流式细胞仪方法结果准确,重复性好,是各种方法中最好的一种,但仪器设备及试剂较昂贵.Background: Eosinophil is an important index for many diseases, several methods to count eosinophil were being used in the laboratories. We investigate the precision and relationship among three methods on detecting eosinophils. Methods: 1. Monoclone antibody with fluorescence CD 16 was labeled to whole blood then measure the negative percentage of the granulocyte with the cytometer; 2. Detecting the eosinophils vvith the SYSMEX SE-9500 blood cell analyzer; 3. Counting the eosinophils using microscope. Results: There were no significant different among the three methods used of detecting eosinophils (P>0.05), but there were significant positive correlation among the methods (r=0.99). Conclusion: It was a heavy and complicated work for the microscopic counting method and the result was not very precise. The operation of SYSMEX SE-9500 blood cell analyzer was simple and it gave the precision result. The cytometry method was better accuracy and highest precision, it was the best among the three methods, but the instrument and reagents were costly.
