摘  要:目的 研究γ-干扰素(IFN-γ)对矽肺大鼠肺中白介素-4(IL-4)和转化生长因子-β1(TGF-β1)蛋白表达的影响。方法 气管内非暴露法给大鼠染石英粉尘20 mg,第2天开始每日注射IFN-γ(1×105IU),分别在染尘后1个月和2个月时每组各处死半数动物,HE和VG染色观察其肺组织病理改变,免疫组化SABC法观察其肺组织IL-4和TGF-β1蛋白表达情况,用图像分析系统进行定量分析。结果 (1)IFN-γ治疗组大鼠的矽肺纤维化程度明显轻于石英对照组,病理分级较石英对照组减轻半级-1级。(2)图像定量分析结果表明,染尘后1个月,石英对照组IL-4阳性细胞积分光密度值为(3.33±1.27)×104,染尘后2个月降至(1.99±0.80)×104,TGF-β1阳性细胞积分光密度值从(3.67±0.63)×104升至(4.90±1.11)× 104;IFN-γ治疗组IL-4阳性细胞积分光密度值从(2.19±0.90)×104降至(0.61±0.22)× 104,TGF-β1阳性细胞积分光密度值从(1.37±0.31)×104升至(1.76±0.72)×104,差异有统计学意义(P<0.05或P<0.01)。在这两个时点,IFN-γ治疗组大鼠肺中IL-4、TGF-β1阳性细胞积分光密度值均低于石英对照组,差异均有统计学意义(P<0.05或P<0.01)。结论 IFN-γ对矽肺大鼠肺中IL-4和TGF-β1的表达具有一定的抑制作用。Objective To study the effect of γ-interferon on the protein expressions of interleukin-4(IL-4) and transforming growth factor-β1(TGF-β1 ) on silicotic rats. Methods Wistar rats were exposed to lower dose(20 mg) of quartz by intratracheal injection.Next day,the experimental rats were treated with γ-interferon( 1 × 105 IU) daily. Rats were killed in 1 month and 2 month to observe the changes of pathology of lung tissue by use of HE and VG staining, and the expressions of IL-4 and TGF-β1 by use of immunohistochemical technique and quantitative analysis of protein expressions by image analysis system. Results (1) The fibrotic degree in silicotic lesions of rat lung was 1/2 - 1 grade less in IFN-γ treated groups than in quartz controls. (2) Quantitative image analysis showed that the integrated optical density ( IOD ) of IL-4 positive cell in quartz controls in 1 month was (3.33 ± 1.27) × 104 and decreased to(1.99 ±0.80) × 104 in 2 month,and that of TGF-β, positive cell increased from (3.67±0.63) × 104 to(4.90± 1.11) × 104;The IOD of IL-4 positive cell in IFN-γ treated groups was decreased from (2.19 ±0.90) × 104 to(0.61 ± 0.22) × 104( P < 0.05),and that of TGF-β1 positive cell increased from (1.37 ±0.31) × 104 to( 1.76 ± 0.72) × 104( P < 0.05) .At these two time points, the IOD of IL-4 and TGF-β1 positive cell in IFN-γ treated groups were lower than those in quartz controls ( P < 0. 05 or P < 0. 01) . Conclusion γ-interferon may inhibit the expressions of IL-4 and TGF-β1 in silicotic rats.
