摘　　要：目的:研究血管紧张素Ⅱ(angiotensinⅡ熏AngⅡ)对血管平滑肌细胞(vascularsmoothmusclecells熏VSMCs)增殖规律及凋亡的影响。方法:用MTT、细胞计数和3H鄄TdR掺入法研究不同浓度和作用时间下AngⅡ对VSMCs增殖的影响;用流式细胞技术研究AngⅡ对VSMCs周期与凋亡的影响;用透射电镜观察AngⅡ作用下VSMCs超微结构的变化。结果:AngⅡ能促进VSMCs增殖,在一定范围内,其作用强度与AngⅡ浓度及作用时间呈正相关;AngⅡ在高浓度(10-4mol/L)时,使VSMCs凋亡率增加;AngⅡ促使细胞周期由G0/G1向S/G2鄄M期转变,使细胞由收缩表型向合成表型转变。结论:AngⅡ对VSMCs的促增殖作用具有剂量和时间依赖性熏大剂量AngⅡ有诱导VSMCs凋亡的倾向。Objective: To study the effects of angiotensinⅡ(AngⅡ) on the proliferation and apoptosis of vascular smooth muscle cells (VSMCs). Methods: The cell proliferation was detected by the methods of MTT, cell number counting and 3H-TdR incorporation, cell cycle and apoptosis ratio were determined by flow cytometry, and cell ultramicrostructure was observed under transmission electron microscope. Results: AngⅡ promoted VSMCs proliferation, and the effect had a positive relation to AngⅡconcentration. It had no effect on VSMCs apoptosis in low and moderate concentrations, but high concentration of AngⅡinduced the apoptosis of VSMCs. AngⅡmade VSMCs transform from G0/G1 to S/G2-M phase, and promoted the transition of VSMCs from contractile to synthetic phenotype. Conclusion: The effect of promoting proliferation on VSMCs of AngⅡ has a concentration- and time-depending style, AngⅡin high concentration induces the apoptosis of VSMCs.
