摘  要:目的 研究γ -干扰素 (IFN γ)对矽肺大鼠肺成纤维生长因子 -b (FGF b)和转化生长因子 - β1(TGF β1)蛋白表达的影响。 方法 采用一次性气管内非暴露法给予雌性Wistar大鼠染尘 (石英粉尘2 0mg/鼠) ,染尘后第 2天开始每天注射IFN γ (1× 10 5IU/鼠 ) ,分别在染尘后 1个月和 2个月时处死动物。病理染色观察实验动物肺组织病理改变 ,免疫组化SABC法观察各组实验动物肺组织FGF b和TGF β1蛋白表达情况 ,并采用图像分析系统对各组蛋白表达进行定量分析。结果 IFN γ治疗组大鼠的矽肺纤维化程度明显轻于石英组。图像定量分析结果表明 ,染尘后 1个月时 ,石英组和治疗组FGF b阳性细胞积分光密度值分别为 (0 93± 0 2 6 )× 10 4,(0 97± 0 19)× 10 4,染尘后 2个月时分别为 (0 89± 0 0 8)× 10 4,(0 80± 0 17)× 10 4,两个时点与对照组比较 ,差异均无显著性。染尘后 1个月至 2个月 ,石英组和IFN γ治疗组TGF β1阳性细胞积分光密度值均呈升高趋势 ,其中治疗组升高有显著意义 (P <0 0 5 )。IFN γ治疗组大鼠肺中TGF β1阳性细胞积分光密度值明显低于石英组 (P <0 0 1)。 结论 FGF b在石英组和治疗组中未见表达增高 ,TGF β1表达增高 ,IFN γ对TGF β1的表达具有抑制作用。Objective To study the effect of γ-interferon on the expression of fibroblast growth factor-b ( FGF-b ) and transform growth factor-β1(TGF-β1) in silicotic lungs of rats.Methods Expose female Wistar rats to lower dosage (20 mg/rat) of quartz by intratracheal injection.The second day after exposure,the experimental rats were treated with γ-interferon (1×10 5IU/rat) per day,rats were sacrificed on the 1st month and 2nd month respectively to observe the pathological changes of silicotic rats by means of HE and VG dyeing,expression of FGF-b and TGF-β1 by means of immunohistochemical technique and quantitative analysis of protein expression by image analysis system.Results The fibrotic degree in silicotic lesions of rat lungs was much milder in IFN-γ treated groups than that of quartz groups.Quantitative image analysis showed that after exposure of one month,integrated optical density(OD) total of FGF-b positive cell in quartz[(0.93±0.26)×10 4]and IFN-γ treated groups[(0.97±0.19)×10 4],and 2 months after exposure[(0.89±0.08)×10 4,(0.80±0.17)×10 4] were not significantly different from that of control group.After 1~2 months exposure,integrated optical density(OD) total of TGF-β1 positive cell in quartz group and IFN-γ treated groups(P<0.05) increased.The integrated OD total of TGF-β1 positive cell in IFN-γ treated groups was lower than those of quartz groups,and the differences were significant(P<0.01).Conclusions The expressions of FGF-b might not be higher in quartz group and IFN-γ treated groups.The expressions of TGF-β1 were higher,and IFN-γ might prevent its expressions.
