摘  要:目的 研究缺氧缺血性脑损伤 (HIBD)后血清和脑组织中NO、NOS的变化 ,探讨其是否存在相关性。 方法 取空白对照组、假手术组、缺氧缺血后 30min、2h、2 4h、72h大鼠血清、脑组织标本进行NO和NOS的检测。NO测定采用硝酸还原酶法 ,用分光光度计比色法测定NO和NOS。 结果  (1)缺氧缺血组NO、NOS增高 ,与假手术组、空白对照组比较差异有显著性 ,但后两组差异无显著性。 (2 )新生大鼠脑缺氧缺血后NO、NOS随时间的延长而逐渐升高。 (3)血清、脑组织NO、NOS的变化存在相关性。 结论 新生大鼠缺氧缺血性脑损伤后血清和脑组织中NO、NOS都有不同程度的升高 ,而且存在相关性。可以通过血NO、NOS的检测了解脑组织的受损情况。Objective To establish the model of hypoxic-ischemic brain damage(HIBD) in neonatal rats and study the relationships among the changes of nitric oxide(NO) and nitric oxide synthase(NOS) in serum and brain tissue of neonatal rats, and to explore the simple and safe method in early diagnosis of hypoxic ischemic encephalopathy(HIE). Methods NO and NOS of serum and brain tissue were determined at 30 minutes 2, 24, 72 hours after damage. 3 groups neonatal rats consisted the control group, the sham group, and the group with HIBD. NO was tested by the nitric acid deoxidize enzyme method, spectrophotometer was used to measure NO and NOS. Results NO and NOS of serum and brain tissue of the group with HIBD were significantly higher than those of control group and sham group. No remarkable different was found between control group and sham group. NO and NOS increased significantly with the lapse of time(serum NO 30 min 1025 μmol/L, 2 h 1409 μmol/L, 24 h 1499 μmol/L and 72 h 1623 μmol/L; NOS 30 min 24.69 U/mL, 2 h 27.16 U/mL, 24 h 19.84 U/mL, 72 h 33.29 {U/mL}), and in brain tissue had equally expression also. Conclusion There is significant correlation between the levels of NO and NOS in serum and brain tissue of neonatal rats with HIBD. The damage of brain can be measured by testing the NO and NOS in blood.
