摘 要:目的了解海南省恶性疟原虫裂殖子表面蛋白 3(pfMSP3)等位基因型的分型特征。方法采用套式PCR法扩增pfMSP3基因的可变区序列 ,对海南省恶性疟原虫分离株进行基因分型 ,并对其代表株的基因片段进行序列分析。结果 4 1份血样扩增出 5 1个pfMSP3基因片段 ,以 3D7型为主导型 (78.4 % ) ,K1型为次要型 (2 1.6 % ) ,其中 ,混合感染率为 2 4 .4 %。序列分析表明 ,海南省恶性疟原虫虫株的 3D7型和K1型的可变区序列与 3D7和K1原型序列具有高度同源性。结论海南省恶性疟原虫的MSP3存在 3D7型和K1型两种等位基因型 ,以Objective To identify the genotypes of merozoite surface protein 3 (MSP3)of Plasmodium falciparum isolates from Hainan province. Methods Nested PCR was applied to amplify the variable regions of MSP3. Results From 41 blood samples from Plasmodium falciparum patients, gene fragments of MSP3 were amplified, of which the 3D7-type allele was dominant (78 4%),followed by K1-type allele (21.6%) . The mixed infection rate of the two different allelic types was 24.4% . Sequence analysis showed that the sequences of 3D7-and K1-type isolates from Hainan Province were highly homologous to that of 3D7 and K1 allelic prototypes respectively. Conclusion Two allelic types of MSP3 gene,3D7-type and K1-type, exist in malaria endemic areas in Hainan province, 3D7-type being dominant.
