摘 要:目的 评价补肾益精方提高绝经后骨质疏松症大鼠模型皮质骨骨质量的有效性 ,探讨本方改善皮质骨力学性能的作用机制。方法 4 0只 12月龄Wistar雌性大鼠随机分为正常对照组 (双侧卵巢假切除术 )、模型对照组 (双侧卵巢切除术 )、补肾益精方组 (双侧卵巢切除术加补肾益精方 ) ,倍美力组 (双侧卵巢切除术加倍美力 ) ,每组 10只 ;给药组的给药时点是双侧卵巢切除术后 90d ,给药组与非给药组的处死时点是卵巢切除术 /假切除术后 180d。测定各组大鼠左股骨中段的截面几何结构 (骨皮质厚度、骨髓腔直径 )、骨矿密度与右股骨三点弯曲的结构 /材料力学参数。结果 模型对照组的截面宏观几何结构参数与正常对照组比较 ,骨髓腔显著扩大 (P <0 .0 5 ) ,骨皮质显著变薄 (P <0 .0 5 ) ;补肾益精方组、倍美力组与模型对照组比较 ,虽然骨髓腔都没有明显缩小 (P >0 .0 5 ) ,但是骨皮质均明显变厚 (P <0 .0 5 )。模型对照组的骨矿密度与正常对照组比较 ,没有明显下降 (P >0 .0 5 ) ;倍美力组与模型对照组比较 ,骨矿密度明显上升 (P <0 .0 5 ) ,补肾益精方组上升不明显 (P >0 .0 5 )。模型对照组的结构力学参数 (最大载荷、结构硬度、结构韧性 )以及材料力学参数(最大应力、弹性模量 )与正常对照组比较 ,均显著下降 (POBJECTIVE To evaluate the effect of the kidney-invigorating prescription on cortical bone of post-menopause osteoporosis rat models. METHOD 40 12-month old female rats were randomly divided into a normal control group (bilateral pseudo-oophorectomy), a model control group (bilateral oophorectomy), a kidney-invigorating group (bilateral oophorectomy) and a Premarin group (bilateral oophorectomy). Different methods of treatment were given to observe the therapeutic effect. RESULT Compared with the normal control group, the cross-section macroscopy geometric structure parameter in the model control group showed marked enlargement of the medullary canal (P<0.05) and marked attenuation of the bone cortex (P<0.05). In the kidney-invigorating group and Premarin group, although medullary canal was not markedly reduced in size (P>0.05), marked thickening of the bone cortex occurred (P<0.05). Compared with the normal control group, the bone density in the model control group did not drop markedly(P>0.05). Compared with the model control group, the bone density in the Premarin group markedly rose (P<0.05) whereas the bone density in the kidney-invigorating group did not rise markedly (P>0.05). Compared with the normal control group, the structural mechanics parameters and material mechanics parameters in the model control group both dropped markedly (P<0.05). Compared with the model control group, the structure hardness and elasticity rose markedly (P<0.05) in the kidney-invigorating group but not markedly in the Premarin group (P>0.05). Other parameters in the two groups also rose (P<0.05). CONCLUSION Kidney-invigorating prescription is effective in increasing the cortical bone quality of post-menopause osteoporosis rat models.
