转化生长因子β_1对软骨样细胞整合素和吞噬细胞糖蛋白-1表达的影响

摘　　要：目的 :检测软骨样细胞 (HEMCSS)α1β1,α2 β1,α5 β13种整合素 (integrins)和吞噬细胞糖蛋白 - 1(CD4 4 )的表达 ,及转化生长因子 β1(TGF - β1)对它们表达水平的影响。方法 :HEMCSS是永生性软骨肉瘤细胞 ,培养于含10 %胎牛血清 ,不含或含 1,5 ,10ng·ml-1的TGF - β1的培养液 ,培养时间 2 4h。共聚焦显微镜观察α1β1,α2 β1,α5 β1和CD4 4在HEMCSS细胞上的表达和分布 ,流式细胞仪检测α1β1,α2 β1,α5 β1和CD4 4在HEMCSS细胞上表达的水平。结果 :共聚焦显微镜结果显示 ,HEMCSS细胞膜表面均有α1β1,α2 β1,α5 β1及CD4 4表达。流式细胞仪检测表明 ,加入TGF - β1,培养HEMCSS细胞 2 4h后可诱导α1β1,α2 β1,α5 β1的表达增加 ,并呈剂量依赖趋势。而 3种不同浓度的TGF - β1培养 2 4h后均减少了HEMCSS细胞CD4 4表达。结论 :TGF - β1可以调节HEMCSS细胞膜上细胞外基质受体的表达 ,从而调节软骨细胞与细胞外基质成分的结合 ,对调节软骨细胞的功能起着重要的作用。Objective: To study the expression of the integrins(α1β1,α2β1,α5β1) and CD44 of the human chondrosarcoma-derived chondrocyte-like cell line(HEMCSS), and study the effects of transforming growth factor β1 on α1β1,α2β1,α5β1 and CD44 expression of HEMCSS. Methods: HEMCSS cells were cultured in the presence of 10% fetal bovine serum, with 0,1, 5, 10 ng·ml -1 TGFβ 1 for 24 h. The expression and location of α1β1,α2β1,α5β1 and CD44 were measured by confocal microscope. The expression level of α1β1,α2β1,α5β1 integrins and CD44 were measured by flow cytometry. Results: HEMCSS cells were found to express α1β1,α2β1,α5β1 and CD44, which were located on the surface of cell membrance by confocal microscope. The expression level of α1β1,α2β1, α5β1 increased in the HEMCSS cells cultured with TGF-β 1 for 24 h, and is concentration-dependent to some degree. However HEMCSS cells were cultured with TGF-β 1 in the three different concentrations groups for 24h, all the expression level of CD44 decreased. Conclusion: TGF-β 1 may regulate the expression of extracellular matrix receptors of HEMCSS cells. This suggests that TGF-β 1 can regulate adhesion of chondrocytes to extracellular matrix proteins, which may play an important role to the function of chondrocytes.
